Abstract

The present invention relates to substituted triazole compounds and compositions comprising substituted triazole compounds. The invention further relates to methods of inhibiting the activity of Hsp90 in a subject in need thereof and methods for preventing or treating hyperproliferative disorders, such as cancer, in a subject in need thereof comprising administering to the subject a substituted triazole compound of the invention, or a composition comprising such a compound.

Claims (70)

1. A method of treating a proliferative disorder in a mammal, comprising administering to the mammal an effective amount of a compound represented by the following structural formula:

or a tautomer, or a pharmaceutically acceptable salt thereof;

wherein:

X 41 is NR 42 ;

X 42 is CR 44 ;

Y 40 is CR 43 ;

Y 41 is CR 45 ;

Y 42 , for each occurrence, is independently C or CR 46 ;

R 7 and R 8 , for each occurrence, is independently, —H, an optionally substituted alkyl, an optionally substituted alkenyl, an optionally substituted alkynyl, an optionally substituted cycloalkyl, an optionally substituted cycloalkenyl, an optionally substituted heterocyclyl, an optionally substituted aryl, an optionally substituted heteroaryl, an optionally substituted aralkyl, or an optionally substituted heteraralkyl;

R 10 and R 11 , for each occurrence, is independently —H, an optionally substituted alkyl, an optionally substituted alkenyl, an optionally substituted alkynyl, an optionally substituted cycloalkyl, an optionally substituted cycloalkenyl, an optionally substituted heterocyclyl, an optionally substituted aryl, an optionally substituted heteroaryl, an optionally substituted aralkyl, or an optionally substituted heteraralkyl; or R 10 and R 11 , taken together with the nitrogen to which they are attached, form an optionally substituted heterocyclyl or an optionally substituted heteroaryl;

R 26 is a lower alkyl;

R 42 is —H, an optionally substituted alkyl, an optionally substituted alkenyl, an optionally substituted alkynyl, an optionally substituted cycloalkyl, an optionally substituted cycloalkenyl, an optionally substituted heterocyclyl, an optionally substituted aryl, an optionally substituted heteroaryl, an optionally substituted aralkyl, an optionally substituted heteraralkyl, hydroxyalkyl, alkoxyalkyl, a haloalkyl, a heteroalkyl, —C(O)R 7 , —(CH 2 ) m C(O)OR 7 , —C(O)OR 7 , —OC(O)R 7 , —C(O)NR 10 R 11 , —S(O) p R 7 , —S(O) p OR 7 , or —S(O) p NR 10 R 11 ;

R 43 and R 44 are, independently, —H, —OH, an optionally substituted alkyl, an optionally substituted alkenyl, an optionally substituted alkynyl, an optionally substituted cycloalkyl, an optionally substituted cycloalkenyl, an optionally substituted heterocyclyl, an optionally substituted aryl, an optionally substituted heteroaryl, an optionally substituted aralkyl, an optionally substituted heteraralkyl, hydroxyalkyl, alkoxyalkyl, halo, cyano, nitro, guanadino, a haloalkyl, a heteroalkyl, —C(O)R 7 , —C(O)OR 7 , —OC(O)R 7 , —C(O)NR 10 R 11 , —NR 8 C(O)R 7 , —SR 7 , —S(O) p R 7 , —OS(O) p R 7 , —S(O) p OR 7 , —NR 8 S(O) p R 7 , —S(O) p NR 10 R 11 , or R 43 and R 44 taken together with the carbon atoms to which they are attached form an optionally substituted cycloalkenyl, an optionally substituted aryl, an optionally substituted heterocyclyl, or an optionally substituted heteroaryl;

R 46 , for each occurrence, is independently selected from the group consisting of H, an optionally substituted alkyl, an optionally substituted alkenyl, an optionally substituted alkynyl, an optionally substituted cycloalkyl, an optionally substituted cycloalkenyl, an optionally substituted heterocyclyl, an optionally substituted aryl, an optionally substituted heteroaryl, an optionally substituted aralkyl, an optionally substituted heteraralkyl, halo, cyano, nitro, guanadino, a haloalkyl, a heteroalkyl, —NR 10 R 11 , —OR 7 , —C(O)R 7 , —C(O)OR 7 , —OC(O)R 7 , —C(O)NR 10 R 11 , —NR 8 C(O)R 7 , —SR 7 , —S(O) p R 7 , —OS(O) p R 7 , —S(O) p OR 7 , —NR 8 S(O) p R 7 , or —S(O) p NR 10 R 11 ;

p, for each occurrence, is independently, 1 or 2; and

m, for each occurrence, is independently 1, 2, 3, or 4.

2. The method of claim 1 , wherein

R 41 is selected from the group consisting of methyl, ethyl, propyl, isopropyl, cyclopropyl, methoxy, ethoxy, propoxy, and cyclopropoxy; and

R 42 is selected from the group consisting of —H, a lower alkyl, a lower cycloalkyl, —C(O)N(R 27 ) 2 , and —C(O)OH, wherein each R 27 is independently —H or a lower alkyl.

3. The method of claim 1 , wherein R 42 is selected from the group consisting of —H, methyl, ethyl, n-propyl, isopropyl, cyclopropyl, n-butyl, sec-butyl, tert-butyl, n-pentyl, n-hexyl, —C(O)OH, —(CH 2 ) m C(O)OH, —CH 2 OCH 3 , —CH 2 CH 2 OCH 3 , and —C(O)N(CH 3 ) 2 .

4. The method of claim 1 , wherein

R 43 and R 44 are, independently, selected from the group consisting of —H, methyl, ethyl, propyl, isopropyl, cyclopropyl, methoxy, ethoxy, propoxy, and cyclopropoxy; or

R 43 and R 44 are taken together with the carbon atoms to which they are attached form a C 5 -C 8 cycloalkenyl or a C 5 -C 8 aryl.

5. The method of claim 1 , wherein R 45 is selected from the group consisting of —H, —OH, —SH, —NH 2 , a lower alkoxy, a lower alkyl amino, and a lower dialkyl amino.

6. The method of claim 5 , wherein R 45 is selected from the group consisting of —H, —OH, methoxy and ethoxy.

7. The method of claim 1 , wherein the compound is represented by the following structural formula:

or a tautomer, or a pharmaceutically acceptable salt thereof.

8. The method of claim 7 , wherein R 43 and R 44 are, independently, selected from the group consisting of —H, methyl, ethyl, propyl, isopropyl, cyclopropyl, methoxy, ethoxy, propoxy, and cyclopropoxy; or

R 43 and R 44 , are taken together with the carbon atoms to which they are attached, form a C 5 -C 8 cycloalkenyl or a C 5 -C 8 aryl.

9. The method of claim 1 , wherein the compound is 5-hydroxy-4-(5-hydroxy-4-(1-methyl-1H-indol-5-yl)-4H-1,2,4-triazol-3-yl)-2-isopropylphenyl dihydrogen phosphate.

10. The method of claim 1 , wherein the proliferative disorder is cancer.

11. The method of claim 10 , wherein the cancer is a c-kit associated cancer, a Bcr-Abl associated cancer, a FLT3 associated cancer or an EGFR associated cancer.

12. The method of claim 1 , wherein the mammal is a human.

13. The method of claim 10 , wherein the mammal is a human.

14. A method of blocking, occluding, or otherwise disrupting blood flow in neovasculature, comprising contacting the neovasculature with an effective amount of a compound represented by the following structural formula:

or a tautomer, or a pharmaceutically acceptable salt thereof;

X 41 is NR 42 ;

X 42 is CR 44 ;

Y 40 is CR 43 ;

Y 41 is CR 45 ;

Y 42 , for each occurrence, is independently C or CR 46 ;

R 7 and R 8 , for each occurrence, is independently, —H, an optionally substituted alkyl, an optionally substituted alkenyl, an optionally substituted alkynyl, an optionally substituted cycloalkyl, an optionally substituted cycloalkenyl, an optionally substituted heterocyclyl, an optionally substituted aryl, an optionally substituted heteroaryl, an optionally substituted aralkyl, or an optionally substituted heteraralkyl;

R 10 and R 11 , for each occurrence, is independently —H, an optionally substituted alkyl, an optionally substituted alkenyl, an optionally substituted alkynyl, an optionally substituted cycloalkyl, an optionally substituted cycloalkenyl, an optionally substituted heterocyclyl, an optionally substituted aryl, an optionally substituted heteroaryl, an optionally substituted aralkyl, or an optionally substituted heteraralkyl; or R 10 and R 11 , taken together with the nitrogen to which they are attached, form an optionally substituted heterocyclyl or an optionally substituted heteroaryl;

R 26 is a lower alkyl;

p, for each occurrence, is independently, 1 or 2; and

m, for each occurrence, is independently 1, 2, 3, or 4.

15. The method of claim 14 , wherein

R 41 is selected from the group consisting of methyl, ethyl, propyl, isopropyl, cyclopropyl, methoxy, ethoxy, propoxy, and cyclopropoxy; and

R 42 is selected from the group consisting of —H, a lower alkyl, a lower cycloalkyl, —C(O)N(R 27 ) 2 , and —C(O)OH, wherein each R 27 is independently —H or a lower alkyl.

16. The method of claim 14 , wherein R 42 is selected from the group consisting of —H, methyl, ethyl, n-propyl, isopropyl, cyclopropyl, n-butyl, sec-butyl, tert-butyl, n-pentyl, n-hexyl, —C(O)OH, —(CH 2 ) m C(O)OH, —CH 2 OCH 3 , —CH 2 CH 2 OCH 3 , and —C(O)N(CH 3 ) 2 .

17. The method of claim 14 , wherein

R 43 and R 44 are, independently, selected from the group consisting of —H, methyl, ethyl, propyl, isopropyl, cyclopropyl, methoxy, ethoxy, propoxy, and cyclopropoxy; or

R 43 and R 44 are taken together with the carbon atoms to which they are attached form a C 5 -C 8 cycloalkenyl or a C 5 -C 8 aryl.

18. The method of claim 14 , wherein R 45 is selected from the group consisting of —H, —OH, —SH, —NH 2 , a lower alkoxy, a lower alkyl amino, and a lower dialkyl amino.

19. The method of claim 18 , wherein R 45 is selected from the group consisting of —H, —OH, methoxy and ethoxy.

20. The method of claim 14 , wherein the compound is represented by the following structural formula:

or a tautomer, or a pharmaceutically acceptable salt thereof.

21. The method of claim 20 , wherein R 43 and R 44 are, independently, selected from the group consisting of —H, methyl, ethyl, propyl, isopropyl, cyclopropyl, methoxy, ethoxy, propoxy, and cyclopropoxy; or

R 43 and R 44 , are taken together with the carbon atoms to which they are attached, form a C 5 -C 8 cycloalkenyl or a C 5 -C 8 aryl.

22. The method of claim 14 , wherein the compound is 5-hydroxy-4-(5-hydroxy-4-(1-methyl-1H-indol-5-yl)-4H-1,2,4-triazol-3-yl)-2-isopropylphenyl dihydrogen phosphate.

23. The method of claim 14 , wherein the neovasculature is in a subject and blood flow in the neovasculature is blocked, occluded, or otherwise disrupted in the subject by administering to the subject an effective amount of the compound.

24. The method of claim 23 , wherein the subject is human.
