Abstract

Provided herein are water-soluble prodrugs, compositions comprising such prodrugs, and related methods of making and administering the same. The prodrugs of the invention comprise a water-soluble polymer having three or more arms, at least three of which are typically covalently attached to an active agent, e.g., a small molecule. The conjugates of the invention provide an optimal balance of polymer size and structure for achieving improved drug loading, since the conjugates of the invention possess three or more active agents releasably attached to a multi-armed water-soluble polymer. The prodrugs of the invention are therapeutically effective, and exhibit improved properties in-vivo when compared to unmodified parent drug.

Claims (18)

1. A pharmaceutical composition comprising:

(i) a pharmaceutically effective amount of a compound having the structure,

or a pharmaceutically acceptable salt form thereof, and

(ii) a pharmaceutically acceptable excipient, wherein

n ranges from 20 to about 300, and the composition is in a form of a sterile lyophilized powder.

2. The pharmaceutical composition of claim 1 , wherein the pharmaceutically acceptable excipient is lactate buffer, and the composition is formulated in a pH range from 3 to 5.

3. The pharmaceutical composition of claim 2 reconstituted in an aqueous vehicle suitable for intravenous infusion.

4. The pharmaceutical composition of claim 3 , wherein the aqueous vehicle is a 5% dextrose solution or a 0.9% sodium chloride solution.

5. The pharmaceutical composition of claim 3 , wherein the composition is reconstituted in a 5% dextrose solution to a final concentration range of compound of approximately 0.12 to 2.8 mg/mL.

6. The pharmaceutical composition of claim 1 , wherein the overall nominal average molecular weight of the compound ranges from about 20,000 to about 80,000 daltons.

7. The pharmaceutical composition of claim 1 , wherein each polymer arm has a molecular weight corresponding to about 5000 daltons.

8. The pharmaceutical composition of claim 1 , further comprising an additional compound having a structure selected from the group consisting of:

or a pharmaceutically acceptable salt form thereof,

wherein O-Irinotecan is

L is the site of attachment, and

n ranges from 20 to 300.

9. The pharmaceutical composition of claim 1 , characterized by an average number of irinotecan molecules per 4-armed polymer in a range from 2.1 to 3.75.

10. The pharmaceutical composition of claim 1 , packaged in a sealed amber glass vial.
