Abstract

The present invention relates to a method for the isolation of proteins that comprise disulfide-bonds in their native conformation. Essentially, a method of the present makes the use of reducing agents such as β-mercaptoethanol or dithiothreitol in protein isolation methods obsolete. A method of the present invention is particularly suitable for the isolation of precursor proteins such as proinsulin from recombinant cells.

Claims (8)

1. A method for isolating a recombinant insulin protein that comprises disulfide-bonds in its native conformation, said method comprising the steps of:

a) maintaining a culture of recombinant yeast host cells containing an expression cassette which directs the expression of the recombinant protein under culture conditions suitable for protein expression;

b) harvesting the host cells from the culture and disrupting the host cells thereby producing a crude cell extract; and

c) isolating the recombinant insulin protein from the cell extract of step b); wherein the headspace above the culture and process liquids in steps b) and c) are flushed with nitrogen gas, and wherein

the recombinant protein isolated in step c) is isolated without the use of reducing agents.

2. A method according to claim 1 , wherein said insulin protein is a recombinant hybrid protein present as inclusion bodies of recombinant cells.

3. A method according to claim 2 , wherein said isolation comprises the harvesting and disruption of said cells, the isolation of said inclusion bodies and/or stabilization of the recombinant insulin protein.

4. A method according to claim 1 , wherein said insulin protein is a precursor protein.
