Abstract

Inhibitors of HCV replication of formula I, including stereochemically isomeric forms, and salts, hydrates, solvates thereof, wherein R and R′ have the meaning as defined herein. The present invention also relates to processes for preparing said compounds, pharmaceutical compositions containing them and their use, alone or in combination with other HCV inhibitors, in HCV therapy.

Claims (31)

1. A compound of Formula I

or a stereoisomer or a tautomer thereof, wherein:

Y is CH, N or CR 4 ;

W is carbonyl, sulfonyl or CR 5 R 6 ;

 independently is selected from a group comprising

R and R′ are independently selected from —CR 1 R 2 R 3 , aryl optionally substituted with 1 or 2 substituents selected from halo and methyl, or heterocycloalkyl, wherein

R 1 is selected from C 1-4 alkyl, C 2-4 a1kyl substituted with methoxy or hydroxyl, and phenyl optionally substituted with 1 or 2 substituents independently selected from halo and methyl;

R 2 is hydroxyl, amino, mono- or di-C 1-4 alkylamino, C 1-4 alkyl- carbonylamino, C 1-4 alkyloxycarbonylamino

R 3 is hydrogen or C 1-4 alkyl;

R 4 is hydrogen, C 1-4 alkyl or Fluoro;

R 5 and R 6 , each independently, are C 1-4 alkyl; or

CR 5 R 6 together form C 3-7 cycloalkyl, oxetane, tetrahydrofurane;

or a pharmaceutically acceptable salt or a solvate thereof.

2. The compound according to claim 1 , which is of formula Ia

wherein A, Y, R and R′ are as defined in claim 1 .

3. The compound according to claim 1 , which is of formula Ib

wherein A, Y, R and R′ are as defined in claim 1 .

4. The compound according to claim 1 , which is of formula Ic

wherein Y, R and R′ are as defined in claim 1 .

5. The compound of claim 1 , wherein each

 independently is selected from a group comprising

6. The compound according to claim 1 wherein at least one

7. The compound according to claim 1 wherein R 2 is C 1-4 alkylcarbonylamino or C 1-4 alkyloxycarbonylamino, and R 3 is hydrogen.

8. The compound according to claim 1 , wherein R 1 is selected from branched C 3-4 alkyl; C 2-3 alkyl substituted with methoxy; and phenyl optionally substituted with 1 substituent selected from halo and methyl.

9. The compound according claim 1 which is of formula Id

wherein Y, R and R′ are as defined in claim 1 .

10. A pharmaceutical composition comprising a compound according to claim 1 and a pharmaceutically acceptable carrier.

11. A method of treating an HCV infection in a mammal in need thereof, comprising administering to said mammal a therapeutically effective amount of a compound of claim 1 .

12. The method of claim 11 , further comprising administration of another HCV inhibitor.

13. The method of claim 12 , wherein said compound of claim 1 and said another HCV inhibitor are administered simultaneously, separately or sequentially.

14. The pharmaceutical composition of claim 10 , further comprising another HCV inhibitor.
