Abstract

Some embodiments are related to a method for determining if a subject has an increased likelihood of high grade dysplasia (HGD) or esophagal adenocarcinoma (EC). An exemplary method can include providing a sample from an AFI patch of the subject's esophagus, assaying the sample for (i) aneuploidy, and (ii) abnormal p53, and (iii) abnormal Cyclin A, wherein if the sample shows the presence of at least two of (i), (ii) and (iii), then the subject is determined as having an increased likelihood of high grade dysplasia (HGD) or esophagal adenocarcinoma (EC). The invention also relates to certain methods of medical treatment.

Claims (16)

1 . A method for determining if a subject has an increased likelihood of high grade dysplasia (HGD) or esophagal adenocarcinoma (EC), the method comprising;

a) providing a sample from an AFI patch of said subject's oesophagus

b) assaying said sample for

(i) aneuploidy, and

(ii) abnormal p53, and

(iii) abnormal Cyclin A

c) wherein if said sample shows the presence of at least two of (i), (ii) and (iii), then said subject is determined as having an increased likelihood of high grade dysplasia (HGD) or esophagal adenocarcinoma (EC).

2 . A method according to claim 1 , wherein said sample is further assayed for one or more of 9p LOH, Runx3, pi 6, 17p LOH, HPP1 or G2.

3 . A method according to claim 1 wherein said sample is a biopsy.

4 . A method according to claim 1 wherein said sample is an endoscopic sample.

5 . A method according to claim 1 wherein aneuploidy is assayed by determining the DNA content by flow cytometry DNA analysis and inferring from said determination whether said DNA content is aneuploid.

6 . A method according to claim 1 wherein p53 is assayed by immunohistochemistry.

7 . A method according to claim 1 wherein Cyclin A is assayed by determining the percentage of surface cells staining positive for Cyclin A.

8 . A method of treatment of a subject comprising determining if a subject has an increased likelihood of high grade dysplasia (HGD) or esophagal adenocarcinoma (EC) according to the method of claim 1 , wherein if said subject is determined as having an increased likelihood of high grade dysplasia (HGD) or esophagal adenocarcinoma (EC) then endoscopic mucosal resection (EMR) and/or radio frequency ablation (RFA) are applied to said subject.

9 . A method according to claim 8 further comprising: applying radio frequency ablation (RFA) to said subject.
