Abstract

Conventional spray-drying methods are improved by incorporation of a pressure nozzle and a diffuser plate to improve the flow of drying gas and a drying chamber extension to increase drying time, such improvements leading to the formation of homogeneous solid dispersions of drugs in concentration-enhancing polymers.

Claims (84)

1. A process for producing a pharmaceutical composition comprising the steps:

(a) forming a feed solution comprising a drug, hydroxypropyl methyl cellulose acetate succinate, and a solvent;

(b) directing said feed solution to a spray-drying apparatus comprising

(i) a drying chamber having a volume V dryer and a height H,

(ii) an atomizer for atomizing said feed solution into droplets, and

(iii) a source of heated drying gas for drying said droplets, said source delivering said drying gas to said drying chamber at a flow rate of G,

wherein V dryer is measured in m 3 ,

H is at least 1.0 m,

G is measured in m 3 /sec,

and wherein the following mathematical relationship is satisfied

(c) atomizing said feed solution into droplets in said drying chamber with the atomizer;

(d) contacting said droplets with said heated drying gas to form particles of a solid amorphous dispersion of said drug homogeneously distributed throughout the hydroxypropyl methyl cellulose acetate succinate; and

(e) collecting said particles.

2. The process of claim 1 wherein said droplets have an average diameter of at least 50 μm and a D 10 of at least 10 μm.

3. The process of claim 1 wherein at least 80 vol % of said particles have diameters of greater than 10 μm.

4. The process of claim 1 wherein at least 90 vol % of said particles have diameters of greater than 10 μm.

5. The process of claim 1 wherein id particles have an average diameter of east 40 μm.

6. The process of claim 1 wherein said particles have an average diameter of at least 50 μm.

7. The process of claim 1 wherein said drying gas is dispersed into said drying chamber such that the primary axis of flow of said drying gas is parallel to the axis of the atomizer.

8. The process of claim 1 wherein the particles of solid amorphous dispersion further comprise a surfactant.

9. A process for producing a pharmaceutical composition comprising the steps:

(a) forming a feed solution comprising a drug, a vinyl copolymer, and a solvent;

(b) directing the feed solution to a spray-drying apparatus comprising

(i) a drying chamber having a volume V dryer and a height H,

(ii) an atomizer for atomizing the feed solution into droplets, and

(iii) a source of heated drying gas for drying the droplets, the source delivering the drying gas to the drying chamber at a flow rate of G,

wherein V dryer is measured in m 3 ,

H is at least 1.0 m,

G is measured in m 3 /sec,

and wherein the following mathematical relationship is satisfied

(c) atomizing the feed solution into droplets in the drying chamber using the atomizer;

(d) contacting the droplets with the heated drying gas to form particles of a solid amorphous dispersion of the drug homogeneously distributed throughout the vinyl copolymer; and

(e) collecting the particles.

10. The process of claim 9 wherein the droplets have an average diameter of at least 50 μm and a D 10 of at least 10 μm.

11. The process of claim 9 wherein at least 80 vol % of the particles have diameters of greater than 10 μm.

12. The process of claim 9 wherein at least 90 vol % of the particles have diameters of greater than 10 μm.

13. The process of claim 9 wherein the particles have an average diameter of at least 40 μm.

14. The process of claim 9 wherein the particles have an average diameter of at least 50 μm.

15. The process of claim 9 wherein the particles of solid amorphous dispersion further comprise a surfactant.

16. A process for producing a pharmaceutical composition comprising the steps:

(a) forming a feed solution comprising a drug, hydroxypropyl methyl cellulose, and a solvent;

(b) directing the feed solution to a spray-drying apparatus comprising

(i) a drying chamber having a volume V dryer and a height H,

(ii) an atomizer for atomizing the feed solution into droplets, and

(iii) a source of heated drying gas for drying the droplets, the source delivering the drying gas to the drying chamber at a flow rate of G,

wherein V dryer is measured in m 3 ,

H is at least 1.0 m,

G is measured in m 3 /sec,

and wherein the following mathematical relationship is satisfied

(c) atomizing the feed solution into droplets in the drying chamber with the atomizer;

(d) contacting the droplets with the heated drying gas to form particles of a solid amorphous dispersion of the drug homogeneously distributed throughout the hydroxypropyl methyl cellulose; and

(e) collecting the particles.

17. The process of claim 16 wherein the droplets have an average diameter of at least 50 μm and a D 10 of at least 10 μm.

18. The process of claim 16 wherein at least 80 vol % of the particles have diameters of greater than 10 μm.

19. The process of claim 16 wherein at least 90 vol % of the particles have diameters of greater than 10 μm.

20. The process of claim 16 wherein the particles have an average diameter of at least 40 μm.

21. The process of claim 16 wherein the particles have an average diameter of at least 50 μm.

22. The process of claim 16 wherein the particles of solid amorphous dispersion further comprise a surfactant.
