Abstract

Novel modulators, including antibodies and derivatives thereof, and methods of using such modulators to treat proliferative disorders are provided.

Claims (30)

1. A method of detecting, diagnosing, or monitoring cancer in a subject, the method comprising the steps of (a) contacting cells of the subject with an anti-DLL3 antibody, wherein the anti-DLL3 antibody comprises residues 24-34 of SEQ ID NO: 96 for CDR-L1, residues 50-56 of SEQ ID NO: 96 for CDR-L2, residues 89-97 of SEQ ID NO: 96 for CDR-L3, residues 31-35 of SEQ ID NO: 97 for CDR-H1, residues 50-65 of SEQ ID NO: 97 for CDR-H2 and residues 95-102 of SEQ ID NO: 97 for CDR-H3, wherein the residues are numbered according to Kabat; and (b) detecting the anti-DLL3 antibody on cancer cells.

2. The method of claim 1 , wherein the cells are in a tissue sample and the contacting is performed in vitro.

3. The method of claim 1 , wherein the contacting is performed in vivo.

4. The method of claim 1 , wherein the cancer cells comprise circulating tumor cells.

5. The method of claim 4 , wherein the circulating tumor cells comprise cancer stem cells.

6. The method of claim 1 , further comprising the step of (c) detecting CD324 on cancer cells.

7. The method of claim 1 , further comprising the step of (c) detecting CD46 on cancer cells.

8. The method of claim 1 , further comprising the step of (c) detecting CD324 and CD46 on cancer cells.

9. The antibody of claim 1 , which is selected from the group consisting of a monoclonal antibody, chimeric antibody, CDR-grafted antibody, humanized antibody, human antibody, primatized antibody, multispecific antibody, bispecific antibody, monovalent antibody, multivalent antibody, anti-idiotypic antibody, diabody, Fab fragment, F(ab′) 2 fragment, Fv fragment, and ScFv fragment; or an immunoreactive fragment thereof.

10. The method of claim 9 , wherein the anti-DLL3 antibody is a murine monoclonal antibody.

11. The method of claim 1 , wherein the anti-DLL3 antibody comprises a light chain variable region comprising an amino acid sequence set forth as SEQ ID NO: 96 and a heavy chain variable region comprising an amino acid sequence set forth as SEQ ID NO: 97.

12. The method of claim 1 , wherein the anti-DLL3 antibody is conjugated to or associated with a reporter molecule.

13. The method of claim 1 , wherein the cancer is lung cancer.

14. The method of claim 13 , wherein the lung cancer is small cell lung cancer.

15. The method of claim 1 , further comprising the step of (c) selecting a subject for treatment using an anti-DLL3 antibody drug conjugate based upon detection of DLL3 on cancer cells.

16. A method of detecting, diagnosing, or monitoring cancer in a subject, the method comprising the steps of (a) contacting a tissue sample from the subject with an anti-DLL3 antibody, wherein the anti-DLL3 antibody comprises residues 24-34 of SEQ ID NO: 96 for CDR-L1, residues 50-56 of SEQ ID NO: 96 for CDR-L2, residues 89-97 of SEQ ID NO: 96 for CDR-L3, residues 31-35 of SEQ ID NO: 97 for CDR-H1, residues 50-65 of SEQ ID NO: 97 for CDR-H2 and residues 95-102 of SEQ ID NO: 97 for CDR-H3, wherein the residues are numbered according to Kabat; and (b) detecting the anti-DLL3 antibody on cancer cells.

17. The method of claim 16 , wherein the anti-DLL3 antibody comprises a murine monoclonal antibody comprising a light chain variable region set forth as SEQ ID NO: 96 and a heavy chain variable region set forth as SEQ ID NO: 97.

18. The method of claim 16 , wherein the anti-DLL3 antibody is conjugated to or associated with a reporter molecule.

19. The method of claim 16 , wherein the cancer is lung cancer.

20. The method of claim 19 , wherein the lung cancer is small cell lung cancer.

21. The method of claim 16 , further comprising the step of (c) selecting a subject for treatment using an anti-DLL3 antibody drug conjugate based upon detection of DLL3 on cancer cells.

22. The method of claim 16 , wherein the tissue sample is chemically fixed.

23. The method of claim 22 , wherein the tissue sample is chemically fixed using formalin.

24. The method of claim 23 , wherein the tissue sample is paraffin embedded.

25. A method of detecting circulating tumor cells, the method comprising the steps of (a) contacting cells with an anti-DLL3 antibody, wherein the anti-DLL3 antibody comprises residues 24-34 of SEQ ID NO: 96 for CDR-L1, residues 50-56 of SEQ ID NO: 96 for CDR-L2, residues 89-97 of SEQ ID NO: 96 for CDR-L3, residues 31-35 of SEQ ID NO: 97 for CDR-H1, residues 50-65 of SEQ ID NO: 97 for CDR-H2 and residues 95-102 of SEQ ID NO: 97 for CDR-H3, wherein the residues are numbered according to Kabat; and (b) detecting the anti-DLL3 antibody on circulating tumor cells.

26. The method of claim 25 , wherein the anti-DLL3 antibody is conjugated to or associated with a reporter molecule.

27. The method of claim 25 , wherein the circulating tumor cells are cancer stem cells.

28. The method of claim 25 , wherein the circulating tumor cells are contained in a patient sample.

29. A method of detecting, diagnosing, or monitoring lung cancer in a subject, the method comprising the steps of (a) contacting a tissue sample from the subject with an anti-DLL3 antibody, wherein the anti-DLL3 antibody comprises residues 24-34 of SEQ ID NO: 96 for CDR-L1, residues 50-56 of SEQ ID NO: 96 for CDR-L2, residues 89-97 of SEQ ID NO: 96 for CDR-L3, residues 31-35 of SEQ ID NO: 97 for CDR-H1, residues 50-65 of SEQ ID NO: 97 for CDR-H2 and residues 95-102 of SEQ ID NO: 97 for CDR-H3, wherein the residues are numbered according to Kabat; and (b) detecting the anti-DLL3 antibody on lung cancer cells.

30. The method of claim 29 , further comprising the step of (c) selecting a subject for treatment using an anti-DLL3 antibody drug conjugate based upon detection of DLL3 on lung cancer cells.
