Fermentation process using specific oxygen uptake rates as a process control
View Patent ↗Specific oxygen uptake (OUR) is used as a process control parameter in fermentation processes. OUR is determined during at least the production phase of a fermentation process, and process parameters are adjusted to maintain the OUR within desired ranges. The invention is particularly applicable when the fermentation is conducted using a microorganism having a natural PDC pathway that has been disrupted so that it no longer functions. Microorganisms of this sort often produce poorly under strictly anaerobic conditions. Microaeration controlled by monitoring OUR allows the performance of the microorganism to be optimized.
1. A fermentation process comprising
a) growing engineered yeast cells of the genus Kluyveromyces in a medium comprising a carbohydrate that the cells are capable of metabolizing, while aerating the medium such that as the cells grow and reproduce the dissolved oxygen tension in the medium is reduced to zero and the cells exhibit a specific oxygen uptake rate (OUR) of at least 10 mmol O 2 /g dry weight of cells/hour (mmol O 2 /gdw/h); and then
b) culturing the cells in a buffered medium under fermentation conditions including microaeration conditions sufficient to provide the culture with a specific oxygen uptake rate of about 0.8 to about 3.0 mmol O 2 /gdw/h.
2. The process of claim 1 , wherein the carbohydrate includes a hexose sugar.
3. The process of claim 2 , wherein the hexose sugar includes glucose.
4. The process of claim 1 , wherein the yeast cells have a disrupted PDC pathway.
