Abstract

A particulate SAE-CD composition is provided. The SAE-CD composition has an advantageous combination of physical properties not found in known solid forms of SAECD. In particular, the SAE-CD composition possesses an advantageous physicochemical and morphological property profile such that it can be tailored to particular uses. The SAE-CD composition of the invention has improved flow and dissolution performance as compared to known compositions of SAE-CD.

Claims (25)

1. A method of preparing a pharmaceutical composition comprising a sulfoalkyl ether cyclodextrin and an active agent, the method comprising

combining a sulfoalkyl ether cyclodextrin starting composition, the active agent, and a liquid carrier to form a solution, and wherein the sulfoalkyl ether cyclodextrin starting composition comprises agglomerated particles and has

a bulk density in the range of about 0.38 g/cm 3 to about 0.66 g/cm 3 ; and

a tapped density in the range of about 0.49 g/cm 3 to about 0.75 g/cm 3 , wherein the liquid carrier is water.

2. The method of claim 1 , wherein the process further comprises drying the solution to form a solid composition.

3. The method of claim 2 , wherein the drying is performed using a fluidized bed spray drying.

4. The method of claim 1 , wherein the molar ratio of the sulfoalkyl ether cyclodextrin to the active agent is in the range of about 10 to about 0.1.

5. The method of claim 1 , wherein the active agent is lamotrigine.

The method of claim 1 , wherein the active agent is selected from antibiotic agent, antihistamine agent, decongestant, anti-inflammatory agent, antiparasitic agent, antiviral agent, local anesthetic, antifungal agent, antibacterial agent, amoebicidal agent, trichomonocidal agent, analgesic agent, anti-arthritic agent, anti-asthmatic agent, anticoagulant agent, anticonvulsant agent, antidepressant agent, antidiabetic agent, antineoplastic agent, anti-psychotic agent, neuroleptic agent, antihypertensive agent, hypnotic agent, sedative agent, anxiolytic energizer agent, anti-Parkinson's disease agent, anti-Alzheimer's disease agent, muscle relaxant agent, antimalarial agent, hormonal agent, contraceptive agent, sympathomimetic agent, hypoglycemic agent, anti-hyperglyceridemia agent, anti-dyslipidemia agent, cholesterol reducing agent, bile acid absorption inhibitor, antilipemic agent, ophthalmic agent, electrolytic agent, diagnostic agent, prokinetic agent, gastric acid secretion inhibitor agent, anti-ulcerant agent, anti-flatulent agent, anti-incontinence agent, cardiovascular agent, corticosteroid, B2 adrenoreceptor agonist, dopamine D2 receptor agonist, anticholinergic agent, IL-5 inhibitor, antisense modulators of IL-5, milrinone lactate, tryptase inhibitor, tachykinin receptor antagonist, leukotriene receptor antagonist, 5-lypoxygenase inhibitor, anti-IgE antibody, protease inhibitor or any combinations thereof.

7. The method of claim 1 , wherein the active agent is selected from food products, nutrients, cosmetics, vitamins, minerals, dietary supplements, sterility inhibitors, fertility instigators, microorganisms, flavoring agents, sweeteners, or any combinations thereof.

8. The method of claim 1 , wherein the active agent is an antioxidant selected from acetone, potassium metabisulfite, potassium sulfite, ascorbic acid, ascorbyl palmitate, citric acid, butylated hydroxyanisole, butylated hydroxytoluene, hypophosphorous acid, monothioglycerol, propyl gallate, sodium ascorbate, sodium citrate, sodium sulfide, sodium sulfite, sodium bisulfite, sodium formaldehyde sulfoxylate, thioglycolic acid, EDTA, pentetate, sodium metabisulfite, or any combinations thereof.

9. A pharmaceutical composition comprising a sulfoalkyl ether cyclodextrin and lamotrigine.

10. The pharmaceutical composition of claim 9 , wherein the composition is a solution and the concentration of the lamotrigine in the solution is about 10 mg/ml.

11. The pharmaceutical composition of claim 9 , wherein the molar ratio of the sulfoalkyl ether cyclodextrin to lamotrigine is in the range of about 10 to about 0.1.

12. The pharmaceutical composition of claim 9 , further comprising one or more excipients.

13. The pharmaceutical composition of claim 9 , further comprising a buffering agent.

14. The pharmaceutical composition of claim 9 , further comprising a flavoring agent.

15. The pharmaceutical composition of claim 9 , wherein the sulfoalkyl ether cyclodextrin starting composition comprises the SAE-CD composition of claim 1 .

16. The composition of claim 9 , wherein the sulfoalkyl ether cyclodextrin is a compound, or mixture thereof, having the structure of Formula I:

wherein:

(a) n is 4, 5, or 6;

(b) R 1 , R 2 , R 3 , R 4 , R 5 , R 6 , R 7 , R 8 , and R 9 are each, independently, —O— or a —O—(C 2-6 alkylene)-SO 3 − group, wherein at least one of R 1 to R 9 is, independently, a —O—(C 2-6 alkylene)-SO 3 − group or a —O—(CH 2 ) m SO 3 − group wherein m is 2 to 6; and

c) S 1 , S 2 , S 3 , S 4 , S 5 , S 6 , S 7 , S 8 , and S 9 , are each, independently, a pharmaceutically acceptable cation.

17. A method of administration, comprising orally administrating the pharmaceutical composition of claim 9 to a subject in need thereof.

A method of administration, comprising administrating the pharmaceutical composition of claim 9 to a subject in need thereof through intrapulmonary, intratracheal, intraaveolar, peroral, ocular, ophthalmic, otic, sublingual, buccal, transdermal, transmucosal, rectal, vaginal, uterine, urethral, intrathecal, nasal, parenteral, intraperitoneal, intramuscular, or subdermal administration.
