Isothiocyanate compounds, pharmaceutical compositions, and uses thereof
Provided herein are compositions of matter and pharmaceutical compositions thereof, for use in inhibiting the growth of various microbial pathogens, including bacteria, fungi, protozoa, and viral pathogens. Also provided herein are methods of treating microbial diseases/infections and cancer with the compositions. The compositions are additionally useful in wood preservation and food preservation by inhibition of microbial growth.
1. A composition of matter according to Formula I:
S═C═N—(R) b —(X) a —(R) c —N═C═S
or a pharmaceutically acceptable salt or thereof, wherein:
a is 1, b is 1, and c is 0 or 1;
X is an aryl or heteroaryl ring system which may be unsubstituted or substituted with from 1 to 4 moieties selected from —Br, —Cl, —OH, —NH 2 , —COOH, OR′—CH 3 or R″—CH 3 wherein R′ and R″, independently, are an alkylene moiety having the formula —(CH 2 ) n —, where n can be an integer from 0 to 20; and
each R, independently, is selected from the group consisting of linear or branched alkylene moieties having from 1 to 20 C atoms; linear or branched alkenylene and alkynylene moieties having from 2 to 20 C atoms; linear or branched heteroalkylene moieties having from 2 to 20 C atoms and from 1 to 3 O, N or S atoms; and heteroarylene, and cycloalkylene moieties having from 5 to 10 C atoms and from 1 to 3 N, S, or O atoms;
provided that R is not —CH 2 — when X is phenyl.
2. The composition of claim 1 , where X is selected from phenyl, biphenyl, napthyl, anthrocyl, indole, pyrimidine, pyrene, furan, and pyridine.
3. The composition of claim 1 , wherein each R, independently, is an alkylene moiety having the formula —(CH 2 ) n —, where n can be an integer from 1 to 20.
4. The composition of claim 2 , wherein the X is a phenyl ring.
5. The composition of claim 1 , wherein R is an heteroalkylene moiety having the formula
—(CH 2 ) 2 —O—(CH 2 ) 2 —O—(CH 2 ) 2 —.
6. The composition of claim 5 , wherein c is 1.
7. The composition of claim 1 , wherein
each R, independently, is selected from the group consisting of linear or branched alkylene moieties having from 1 to 20 C atoms and linear or branched heteroalkylene moieties having from 2 to 20 C atoms and from 1 to 3 O, N or S atoms.
8. The composition of claim 1 , wherein
each R, independently, is linear or branched alkylene moieties having from 1 to 20 C atoms.
9. The composition of claim 1 , wherein
each R, independently, is linear or branched heteroalkylene moieties having from 2 to 20 C atoms and from 1 to 3 O, N or S atoms.
10. A composition of matter according to Formula I:
S═C═N—(R) b —(X) a —(R) c —N═C═S
or a pharmaceutically acceptable salt or thereof, wherein:
a and c are independently 0 or 1, b is 1;
X is an aryl or heteroaryl ring system which may be unsubstituted or substituted with from 1 to 4 moieties selected from —Br, —Cl, —OH, —NH 2 , —COOH, OR′—CH 3 or R″—CH 3 wherein R′ and R″, independently, are an alkylene moiety having the formula —(CH 2 ) n —, where n can be an integer from 0 to 20; and
each R, independently, is selected from the group consisting of linear or branched alkylene moieties having from 7 to 20 C atoms; linear or branched alkenylene and alkynylene moieties having from 2 to 20 C atoms; and linear or branched heteroalkylene moieties having from 2 to 20 C atoms and from 1 to 3 O, N or S atoms.
11. The composition of claim 10 , wherein a and c are 0, and R is an heteroalkylene moiety having the formula
—(CH 2 ) 2 —O—(CH 2 ) 2 —O—(CH 2 ) 2 —.
12. The composition of claim 10 , wherein a and c are 0, and R is an linear alkylene having from 7 C atoms.
13. The composition of claim 10 , wherein a and c are 0, and R is an linear alkylene having from 8 C atoms.
14. A composition of matter according to Formula I:
S═C═N—(R) b —(X) a —(R) c —N═C═S
or a pharmaceutically acceptable salt or thereof, wherein:
a, b, and c are each 1;
X is an aryl or heteroaryl ring system which may be unsubstituted or substituted with from 1 to 4 moieties selected from —Br, —Cl, —OH, —NH 2 , —COOH, OR′—CH 3 or R″—CH 3 wherein R′ and R″, independently, are an alkylene moiety having the formula —(CH 2 ) n —, where n can be an integer from 0 to 20; and
each R, independently, is selected from the group consisting of linear or branched alkylene moieties having from 1 to 20 C atoms; linear or branched alkenylene and alkynylene moieties having from 2 to 20 C atoms; linear or branched heteroalkylene moieties having from 2 to 20 C atoms and from 1 to 3 O, N or S atoms; and arylene, heteroarylene, and cycloalkylene moieties having from 5 to 10 C atoms and from 1 to 3 N, S, or O atoms.
15. The composition of claim 14 , wherein each R is independently a linear or branched alkylene moiety having from 1 to 20 C atoms, and X is an aryl ring system which may be unsubstituted or substituted with from 1 to 4 moieties selected from —Br, —Cl, —OH, —NH2, —COOH, OR′—CH3 or R″—CH3 wherein R′ and R″, independently, are an alkylene moiety having the formula —(CH2)n-, where n can be an integer from 0 to 20.
16. The composition of claim 14 , wherein X is phenyl.
17. The composition of claim 14 , wherein X is phenyl and R is CH 2 .
