Abstract

A mutant of a potentially therapeutic anti-CD40 antibody is provided which mutant has reduced ADCC and CDC activities designed to be optimized as a pharmaceutical agent. A mutant of an agonistic anti-CD40 antibody, comprising mutation and/or substitution of at least one amino acid in the constant region to reduce the ADCC and/or CDC activities therein, and a mutant of an antagonistic anti-CD40 antibody, comprising at least one mutation or substitution in the constant region to reduce the ADCC and/or CDC activities therein, both mutants having at least a hinge region derived from a human IgG2.

Claims (10)

1. An antibody prepared by a process comprising:

(i) culturing an animal cell containing (a) a nucleic acid that encodes a heavy chain consisting of a heavy chain signal sequence and an amino acid sequence consisting of Q at position 27 to K at position 474 of SEQ ID NO: 140 and (b) a nucleic acid that encodes a light chain consisting of a light chain signal sequence and an amino acid sequence consisting of A at position 23 to C at position 235 of SEQ ID NO: 142 to produce an antibody that binds human CD40; and

(ii) purifying the antibody,

wherein the purified antibody has substantially no agonistic activity.

2. The antibody of claim 1 , wherein the heavy chain is produced by removing the signal sequence from a polypeptide represented by SEQ ID NO: 140.

3. The antibody of claim 1 , wherein the heavy chain comprises the amino acid sequence set forth in SEQ ID NO: 46.

4. The antibody of claim 1 , wherein the heavy chain comprises a variable region consisting of amino acid residues 27 to 147 of SEQ ID NO: 46.

5. The antibody of claim 1 , wherein the light chain is produced by removing the signal sequence from a polypeptide represented by SEQ ID NO: 142.

6. The antibody of claim 1 , wherein the light chain comprises the amino acid sequence set forth in SEQ ID NO: 48.

7. The antibody of claim 1 , wherein the light chain comprises a variable region consisting of amino acid residues 23 to 128 of SEQ ID NO: 48.
