Abstract

The present invention provides thermostable enzymes, such as DNA polymerases and restriction endonucleases, that are substantially free from contamination with nucleic acids. The invention also provides methods for the production of these enzymes, and kits comprising these enzymes which may be used in amplifying or sequencing nucleic acid molecules, including through use of the polymerase chain reaction (PCR).

Claims (10)

1. An active thermostable enzyme preparation that is substantially free of nucleic acids as determined by an assay for measuring incorporation of a nucleotide in the absence of an exogenous nucleic acid template.

2. The enzyme preparation of claim 1 , wherein said nucleotide is 3 H-labeled.

3. The enzyme preparation of claim 1 , wherein said thermostable enzyme is a DNA polymerase enzyme.

4. The enzyme preparation of claim 3 , wherein said DNA polymerase enzyme is Taq DNA polymerase, Tne DNA polymerase, Tma DNA polymerase, or a mutant, derivative or fragment thereof having DNA polymerase activity.

5. The enzyme preparation of claim 1 , wherein said enzyme preparation is stable at temperatures from about −80° C. to 25° C.

6. The enzyme preparation of claim 1 , wherein said enzyme preparation is in lyophilized form.

7. A composition comprising an active thermostable enzyme preparation that is substantially free of nucleic acids as determined by an assay for measuring incorporation of a nucleotide in the absence of an exogenous nucleic acid template.

8. The composition of claim 7 , wherein said nucleotide is 3H-labeled.

9. The composition of claim 7 , wherein said thermostable enzyme is a DNA polymerase enzyme.

10. The composition of claim 9 , wherein said DNA polymerase enzyme is Taq DNA polymerase, Tne DNA polymerase, Tma DNA polymerase, or a mutant, derivative or fragment thereof having DNA polymerase activity.
