Abstract

The present invention provide purified Flt4 receptor tyrosine kinase polypeptides and fragments thereof, polynucleotides encoding such polypeptides, antibodies that specifically bind such polypeptides, and uses therefor.

Claims (40)

1. A polynucleotide comprising a nucleotide sequence that encodes a fusion protein comprising:

(a) a polypeptide comprising a portion of a human Vascular Endothelial Growth Factor Receptor-3 (VEGFR-3) extracellular domain (EC); wherein said portion and said polypeptide bind to VEGFR-3 ligands VEGF-C and VEGF-D and comprise at least the first, second, and third immunoglobulin-like domains of the VEGFR-3-EC, wherein the polypeptide lacks VEGFR-3-EC immunoglobulin-like domains IV-VII; and

(b) an immunoglobulin-like domain of human VEGFR-2 or an immunoglobulin Fc peptide,

wherein (a) is fused to (b), and

wherein the fusion protein binds VEGF-C and VEGF-D, is soluble, and lacks any transmembrane domain.

2. The polynucleotide of claim 1 , wherein the portion of the VEGFR-3-EC consists of the first, second, and third immunoglobulin-like domains of the VEGFR-3-EC.

3. The polynucleotide according to claim 1 , wherein the portion of the VEGFR-3-EC consists of the first through third immunoglobulin-like domains of SEQ ID NO: 2.

4. The polynucleotide of claim 1 , wherein the polypeptide comprises amino acids 47-314 of SEQ ID NO: 2.

5. The polynucleotide of claim 1 , wherein the polypeptide in the fusion protein is fused to an immunoglobulin-like domain of human VEGFR-2.

6. The polynucleotide of claim 1 , wherein the polypeptide in the fusion protein is fused to an immunoglobulin Fc peptide.

7. The polynucleotide according to claim 1 , further comprising an expression control sequence operatively linked to the sequence that encodes the fusion protein.

8. An expression vector comprising an expression control sequence operatively linked to the polynucleotide according to claim 1 .

9. The expression vector according to claim 8 , wherein the expression control sequence comprises a promoter that promotes expression in a mammalian cell.

10. The expression vector according to claim 9 that is a viral vector selected from the group consisting of retrovirus, adenovirus, adeno-associated virus, vaccinia virus, and herpesvirus vectors.

11. A host cell transformed or transfected with a vector according to claim 9 .

12. The polynucleotide according to claim 1 , wherein the encoded fusion protein is capable of inhibiting at least one of: VEGF-C-mediated stimulation of endothelial cells that express VEGFR-3; and VEGF-C-induced phosphorylation of VEGFR-3.

13. The polynucleotide according to claim 1 , wherein the encoded fusion protein is capable of inhibiting VEGF-C-induced and VEGF-D-induced signaling of VEGFR-3.

A polynucleotide comprising a nucleotide sequence that encodes a fusion protein comprising a polypeptide consisting essentially of: (i) a fragment of human Vascular Endothelial Growth Factor Receptor-3 (VEGFR-3) consisting essentially of the first through third immunoglobulin-like domains of human VEGFR-3 extracellular domain (EC), and (ii) an immunoglobulin Fc peptide,

wherein (i) is fused to (ii), and

wherein the polypeptide binds both VEGF-C and VEGF-D.

15. The polynucleotide according to claim 14 , wherein the fragment comprises amino acids 47-314 of SEQ ID NO: 2.

16. The polynucleotide according to claim 14 , further comprising an expression control sequence operatively linked to the sequence that encodes the fusion protein.

17. An expression vector comprising an expression control sequence operatively linked to the polynucleotide according to claim 14 .

18. The expression vector according to claim 17 , wherein the expression control sequence comprises a promoter that promotes expression in a mammalian cell.

19. The expression vector according to claim 18 that is a viral vector selected from the group consisting of retrovirus, adenovirus, adeno-associated virus, vaccinia virus, and herpesvirus vectors.

20. A host cell transformed or transfected with a vector according to claim 18 .

21. The polynucleotide according to claim 14 , wherein the encoded fusion protein is capable of inhibiting at least one of: VEGF-C-mediated stimulation of endothelial cells that express VEGFR-3; and VEGF-C-induced phosphorylation of VEGFR-3.

22. The polynucleotide according to claim 14 , wherein the encoded fusion protein is capable of inhibiting VEGF-C-induced and VEGF-D-induced signaling of VEGFR-3.

23. A polynucleotide comprising a nucleotide sequence that encodes a chimeric polypeptide comprising a fragment of a human Vascular Endothelial Growth Factor Receptor-3 (VEGFR-3) extracellular domain (EC) fused to a heterologous polypeptide;

wherein said fragment binds to VEGF-C and VEGF-D and comprises at least the first through third immunoglobulin-like domains of the VEGFR-3-EC, wherein the polypeptide lacks VEGFR-3-EC immunoglobulin-like domains IV-VII; and wherein the polypeptide is a soluble polypeptide lacking any transmembrane domain.

24. The polynucleotide according to claim 23 , wherein the heterologous polypeptide increases serum solubility or serum half-life of the fragment of human VEGFR-3-EC.

25. The polynucleotide according to claim 23 , wherein the fragment comprises amino acids 47-314 of SEQ ID NO: 2.

26. The polynucleotide of claim 23 , wherein the heterologous polypeptide is an immunoglobulin Fc peptide.

27. The polynucleotide according to claim 23 , further comprising an expression control sequence operatively linked to the sequence that encodes the chimeric polypeptide.

28. An expression vector comprising an expression control sequence operatively linked to the polynucleotide according to claim 23 .

29. The expression vector according to claim 28 , wherein the expression control sequence comprises a promoter that promotes expression in a mammalian cell.

30. The expression vector according to claim 29 that is a viral vector selected from the group consisting of retrovirus, adenovirus, adeno-associated virus, vaccinia virus, and herpesvirus vectors.

31. A host cell transformed or transfected with a vector according to claim 29 .

32. The polynucleotide according to claim 23 , wherein the encoded chimeric polypeptide is capable of inhibiting at least one of: VEGF-C-mediated stimulation of endothelial cells that express VEGFR-3; and VEGF-C-induced phosphorylation of VEGFR-3.

33. The polynucleotide according to claim 23 , wherein the encoded chimeric polypeptide is capable of inhibiting VEGF-C-induced and VEGF-D-induced signaling of VEGFR-3.
