Abstract

Methods for the safe administration of imidazole or imidazolium compounds, and conditions that may be treated by these methods, are described herein.

Claims (37)

1. A method of safely and effectively delivering zoledronic acid to the blood through repeated oral administration comprising:

mixing zoledronic acid with a diluent, binder, disintegrating agent, or lubricant to form a pharmaceutical composition;

forming the pharmaceutical composition into a pill, tablet, or capsule containing at least 10% zoledronic acid by weight, and containing about 30 mg to about 300 mg of zoledronic acid in the acid or a sodium salt form; and

orally administering the pill, tablet, or capsule to a human being at least twice a month and no more frequently than once a day.

2. The method of claim 1 , wherein, over a period of a month, a total of about 30 mg to about 300 mg of oral zoledronic acid is administered to the human being in two or more individual doses.

3. The method of claim 1 , wherein the pill, tablet, or capsule containing zoledronic acid is orally administered for about 3 to about 10 consecutive days.

4. The method of claim 2 , wherein zoledronic acid is administered in a manner and amount that results in the human being having an AUC 0-24 of zoledronic acid that is about 100 ng·h/mL to about 500 ng·h/mL with each administration of zoledronic acid.

5. The method of claim 2 , wherein zoledronic acid is administered in a manner and amount that results in the human being having an AUC 0-24 of zoledronic acid that is about 50 ng·h/mL to about 500 ng·h/mL with each administration of zoledronic acid.

6. The method of claim 2 , wherein the pill, tablet, or capsule contains about 30 mg to about 60 mg of zoledronic acid equivalent based upon the weight of the acid form.

7. The method of claim 2 , wherein the pill, tablet, or capsule contains about 50 mg of zoledronic acid equivalent based upon the weight of the acid form.

8. The method of claim 2 , wherein the zoledronic acid is administered in a manner that results in an oral bioavailability of zoledronic acid of 1.2% to 4%.

9. The method of claim 8 , wherein the pill, tablet, or capsule contains about 40 mg to about 60 mg of zoledronic acid equivalent based upon the weight of the acid form.

10. The method of claim 8 , wherein the pill, tablet, or capsule contains about 50 mg of zoledronic acid equivalent based upon the weight of the acid form.

11. The method of claim 1 , wherein the pill, tablet, or capsule containing zoledronic acid is orally administered to the human being once a week for at least two weeks.

12. The method of claim 11 , wherein the pill, tablet, or capsule containing zoledronic acid is orally administered for about 3 consecutive weeks to about 10 consecutive weeks.

13. The method of claim 11 , wherein zoledronic acid is administered in a manner and amount that results in the human being having an AUC 0-24 of zoledronic acid that is about 50 ng·h/mL to about 500 ng·h/mL with each administration of zoledronic acid.

14. The method of claim 11 , wherein zoledronic acid is administered in a manner and amount that results in the human being having an AUC 0-24 of zoledronic acid that is about 100 ng·h/mL to about 500 ng·h/mL with each administration of zoledronic acid.

15. The method of claim 11 , wherein the pill, tablet, or capsule contains about 30 mg to about 60 mg of zoledronic acid equivalent based upon the weight of the acid form.

16. The method of claim 11 , wherein the pill, tablet, or capsule contains about 50 mg of zoledronic acid equivalent based upon the weight of the acid form.

17. The method of claim 3 , wherein the zoledronic acid is administered in a manner that results in an oral bioavailability of zoledronic acid of 1.1% to 4%.

18. The method of claim 17 , wherein the pill, tablet, or capsule contains about 40 mg to about 60 mg of zoledronic acid equivalent based upon the weight of the acid form.

19. The method of claim 17 , wherein the pill, tablet, or capsule contains about 50 mg of zoledronic acid equivalent based upon the weight of the acid form.

20. The method of claim 1 , wherein the pill, tablet, or capsule comprises more than about 20% zoledronic acid by weight.

21. A method of safely and effectively delivering zoledronic acid to the blood through repeated oral administration comprising:

mixing zoledronic acid with a diluent, binder, disintegrating agent, or lubricant to form a pharmaceutical composition;

forming the pharmaceutical composition into a pill, tablet, or capsule containing at least 10% zoledronic acid by weight, and containing about 100 mg to about 250 mg of zoledronic acid;

obtaining approval from the U.S. Food and Drug Administration to use the pill, tablet, or capsule in a pain indication that is not associated with osteoporosis or cancer; and

orally administering the pill, tablet, or capsule to a human being at least twice a month and no more frequently than once a day.

22. The method of claim 21 , wherein the pill, tablet, or capsule containing zoledronic acid is orally administered two to five times within a period of a month.

23. The method of claim 21 , wherein the pill, tablet, or capsule contains about 150 mg to about 200 mg of zoledronic acid equivalent based upon the weight of the acid form.

24. The method of claim 21 , wherein the pill, tablet, or capsule contains about 200 mg to about 250 mg of zoledronic acid equivalent based upon the weight of the acid form.

25. The method of claim 21 , wherein zoledronic acid is administered in a manner and amount that results in the human being having an AUC 0-24 of zoledronic acid that is about 100 ng·h/mL to about 500 ng·h/mL with each administration of zoledronic acid.

26. The method of claim 21 , wherein zoledronic acid is administered in a manner and amount that results in the human being having an AUC 0-24 of zoledronic acid that is about 50 ng·h/mL to about 500 ng·h/mL with each administration of zoledronic acid.

27. The method of claim 21 , wherein the zoledronic acid is administered in a manner that results in an oral bioavailability of zoledronic acid of 1% to about 4%.

28. The method of claim 21 , wherein, over a period of a month, at least about 300 mg of oral zoledronic acid is administered to the human being in two or more individual doses.

29. The method of claim 21 , wherein, over a period of six months, at least about 400 mg of oral zoledronic acid is administered to the human being in two or more individual doses.

30. The method of claim 21 , wherein the pill, tablet, or capsule contains zoledronic acid in a salt form.
