View Patent ↗Disclosed is a cell wall C-polysaccharide antigen of Streptococcus pneumoniae which contains not more than 10% by weight of protein, and preferably less which has been purified with 0.1N Na OH prior to deproteinizing. Also disclosed are polyvalent antibodies raised against Streptococcus pneumoniae which have been affinity purified by passing them over a chromatographic affinity matrix to which is coupled the purified and at least partially deproteinized antigen to render them antigen-specific.
1. A method for determining the presence of Streptococcus pneumoniae in a liquid sample, the method comprising:
(a) applying the liquid sample to a porous test strip, wherein the porous test strip defines a liquid flow path, the porous test strip comprising:
(i) a sample receiving zone for receiving the liquid sample;
(ii) multiple binding agents disposed in a dried state at a binding zone along the flow path of the test strip downstream from the sample receiving zone, each binding agent comprising:
a) a detectable particle and
b) polyclonal antibodies affinity purified with isolated and purified C-polysaccharide antigen of Streptococcus pneumoniae , which specifically bind to a cell wall C-polysaccharide antigen of Streptococcus pneumoniae preparation, said preparation has not more than about 10% by weight of protein,
wherein the binding agents are essentially free of antibodies that bind proteins of a C-polysaccharide antigen of Streptococcus pneumoniae;
(iii) multiple capture agents disposed in a dried state at a capture zone, each capture agent comprising the same affinity-purified polyclonal antibodies in unconjugated form; and
(iv) said capture zone downstream from the sample receiving zone; and
(b) allowing said sample to flow laterally along the flow path of the test strip, mobilizing the binding agents;
(c) binding, in the presence of the C-polysaccharide antigen of Streptococcus pneumoniae , a complex comprising one of the binding agents bound to the antigen in the capture zone, wherein the formation of a colored line is indicative of the presence of Streptococcus pneumoniae in the liquid sample.
2. The method according to claim 1 , wherein the cell wall C-polysaccharide antigen is indigenous to all serotypes of Streptococcus pneumoniae.
3. The method of claim 1 , wherein the liquid sample is urine.
4. The method according to claim 1 , in which the antigen-specific antibodies are present in a concentration of between 7.7 nanograms/mm2 of surface area and 385 nanograms/mm2 of surface area at each site of a test strip at which antigen:antibody reaction is to occur.
