Abstract

The invention provides a method of treating T-cell mediated diseases and a method of inhibiting the activation of T-cells using certain diketopiperazines. The invention also provides methods of synthesizing diketopiperazines and pharmaceutical compositions comprising certain diketopiperazines. The invention further provides methods of making improved pharmaceutical compositions of proteins and peptides by either increasing or decreasing the content of diketopiperazines in the compositions and the resultant improved pharmaceutical compositions.

Claims (13)

1. A pharmaceutical composition comprising a filtrate produced by passing a solution of albumin over an ultrafiltration membrane with a molecular weight cutoff that retains albumin, wherein the filtrate comprises an alanine-aspartic acid diketopiperazine (DA-DKP) and a component selected from the group consisting of caprylic acid, N-acetyl tryptophan and combinations thereof, wherein the composition is an isotonic aqueous solution, and wherein the composition is formulated for administration as an eye drop.

2. The composition of claim 1 , wherein the composition further comprises an aqueous or nonaqueous base and a component selected from the group consisting of dispersing agents, solubilizing agents and suspending agents.

3. The composition of claim 1 wherein the filtrate contains components having molecular weights less than 3000.

4. The composition of claim 1 wherein the albumin is a human albumin.

5. A pharmaceutical product comprising a plastic bottle having a shaped closure adapted to deliver liquid contents dropwise, wherein the plastic bottle comprises a filtrate produced by passing a solution of albumin over an ultrafiltration membrane with a molecular weight cutoff that retains albumin, wherein the filtrate comprises a pharmaceutical composition comprising an alanine-aspartic acid diketopiperazine (DA-DKP) and a component selected from the group consisting of caprylic acid, N-acetyl tryptophan and combinations thereof, wherein the composition is an isotonic aqueous solution.

6. The pharmaceutical product of claim 5 , wherein the composition further comprises an aqueous or nonaqueous base and a component selected from the group consisting of dispersing agents, solubilizing agents and suspending agents.

7. The pharmaceutical product of claim 5 , wherein the filtrate contains components having molecular weights less than 3000.

8. The pharmaceutical product of claim 5 , wherein the albumin is a human albumin.

9. A method of inhibiting ophthalmic inflammation, comprising administering to an animal in need thereof a pharmaceutical composition comprising a filtrate produced by passing a solution of albumin over an ultrafiltration membrane with a molecular weight cutoff that retains albumin, wherein the filtrate comprises an alanine-aspartic acid diketopiperazine (DA-DKP) and a component selected from the group consisting of caprylic acid, N-acetyl tryptophan and combinations thereof, wherein the composition is an isotonic aqueous solution, and wherein the composition is formulated for administration as an eye drop.

10. The method of claim 9 , wherein the animal has an inflammatory eye condition.

11. The method of claim 9 , wherein the composition further comprises an aqueous or nonaqueous base and a component selected from the group consisting of dispersing agents, solubilizing agents and suspending agents.

12. The method of claim 9 , wherein the filtrate contains components having molecular weights less than 3000.

13. The method of claim 9 , wherein the albumin is a human albumin.
