Abstract

Strains of cyanobacteria that produce high levels of alpha ketoglutarate (AKG) and pyruvate are disclosed herein. Methods of culturing these cyanobacteria to produce AKG or pyruvate and recover AKG or pyruvate from the culture are also described herein. Nucleic acid sequences encoding polypeptides that function as ethylene-forming enzymes and their use in the production of ethylene are further disclosed herein. These nucleic acids may be expressed in hosts such as cyanobacteria, which in turn may be cultured to produce ethylene.

Claims (21)

1. An isolated nucleic acid molecule encoding a polypeptide that functions as an ethylene-forming enzyme, wherein the nucleic acid molecule has a sequence at least 90% identical to SEQ ID NO:3.

2. The isolated nucleic acid molecule of claim 1 , wherein the nucleic acid molecule has a sequence at least 95% identical to SEQ ID NO:3.

3. The isolated nucleic acid molecule of claim 1 , wherein the nucleic acid molecule has the sequence of SEQ ID NO:3.

4. The isolated nucleic acid molecule of claim 1 , further comprising a promoter operably linked to the nucleic acid molecule.

5. The isolated nucleic acid molecule of claim 4 , wherein the promoter is petE or psbA.

6. An expression vector comprising the nucleic acid molecule of claim 1 .

7. A host cell comprising the expression vector of claim 6 .

8. The host cell of claim 7 , wherein the cell is a microbial cell.

9. The host cell of claim 7 , wherein the cell is a cyanobacterial cell.

10. The host cell of claim 7 , wherein the cell is a Synechocystis cell.

11. The host cell of claim 7 , wherein the cell is a Synechocystis sp. PCC6803 cell.

12. A method for producing ethylene, comprising:

a) culturing a host cell that expresses a recombinant ethylene-forming enzyme under conditions that allow for ethylene production, wherein the recombinant ethylene-forming enzyme is encoded by a nucleic acid molecule with a sequence at least 90% identical to SEQ ID NO:3; and

b) isolating ethylene from the culture.

13. The method of claim 12 , wherein the cell is a Synechocystis cell.

14. The method of claim 12 , further comprising a step of replenishing components of a culture medium that are depleted during the culturing step to the cell culture.

15. The method of claim 12 , wherein ethylene is produced at a peak production rate of at least 500 nL mL −1 hr −1 .

16. The method of claim 12 , wherein the step of culturing comprises providing the cell carbon dioxide and light.

17. The method of claim 16 , wherein the light is sunlight.

18. The method of claim 16 , wherein the carbon dioxide is atmospheric carbon dioxide.

19. A method for producing ethylene, comprising culturing the host cell of claim 7 under conditions that allow for the production of ethylene by the cell and isolating the ethylene produced by the cell.
