Abstract

A technique relates sorting biopolymers. The biopolymers are introduced into a nanopillar array, and the biopolymers include a first population and a second population. The nanopillar array includes nanopillars arranged to have a gap separating one from another. The biopolymers are sorted through the nanopillar array by transporting the first population of the biopolymers less than a predetermined bumping size according to a fluid flow direction and by transporting the second population of the biopolymers at least the predetermined bumping size according to a bumped direction different from the fluid flow direction. The nanopillar array is configured to employ the gap with a gap size less than 300 nanometers in order to sort the biopolymers.

Claims (24)

1. A method of sorting biopolymers, the method comprising:

providing a nanopillar array having a substrate material that has a non-uniform gap size between nanopillars, the nanopillars having a coating of a different material from the substrate material that provides a uniform gap size, wherein the uniform gap size is about or below 100 nanometers;

introducing the biopolymers into the nanopillar array, the biopolymers including a first population and a second population; and

sorting the biopolymers through the nanopillar array by transporting the first population of the biopolymers less than a predetermined bumping size according to a fluid flow direction and by transporting the second population of the biopolymers at least the predetermined bumping size according to a bumped direction different from the fluid flow direction.

2. The method of claim 1 , wherein the biopolymers include DNA and RNA.

3. The method of claim 1 , wherein the first population of the biopolymers when uncoiled are shorter than the second population of biopolymers when uncoiled.

4. The method of claim 1 , wherein the first population of the biopolymers includes lengths of at least one of 1 kilo base pairs and 2 kilo base pairs.

5. The method of claim 1 , wherein the second population of the biopolymers includes lengths at least 5 kilo base pairs or greater.

6. The method of claim 1 , wherein the second population of the biopolymers includes lengths in a range of about 5 kilo base pairs to about 166 kilo base pairs.

7. The method of claim 1 , wherein the first population and the second population of biopolymers are sorted when coiled and uncoiled through the nanopillar array.

8. The method of claim 1 , wherein the uniform gap size is less than 100 nanometers.

9. A method of separating and identifying unknown biopolymers, the method comprising:

providing a nanopillar array having a substrate material that has a non-uniform gap size between nanopillars, the nanopillars having a coating of a different material from the substrate material that provides a uniform gap size, wherein the uniform gap size is about or below 100 nanometers;

introducing unknown biopolymers and biopolymer-binding proteins into the nanopillar array, wherein when the unknown biopolymers bind with the biopolymer-binding proteins, biopolymer-protein complexes are formed, and wherein when the unknown biopolymers do not bind with the biopolymer-binding proteins, the unknown biopolymers and biopolymer-binding proteins remain unbound;

sorting the unknown biopolymers and the biopolymer-binding proteins through the nanopillar array by transporting any of the unknown biopolymers and the biopolymer-binding proteins which are unbound in a fluid flow direction, as the unknown biopolymers and the biopolymer-binding proteins which are unbound are less than a predetermined bumping size;

sorting the unknown biopolymers and the biopolymer-binding proteins through the nanopillar array by transporting the biopolymer-protein complexes in a bumped direction, as the biopolymer-protein complexes are at least the predetermined bumping size.

10. The method of claim 9 , wherein the biopolymers include DNA and RNA; and

wherein the DNA and the RNA can form DNA-protein complex and RNA-protein complex, respectively, with specific protein molecules of the biopolymer-binding proteins.

11. The method of claim 9 , wherein the sorting is utilized to identify the unknown biopolymers using the biopolymer-binding proteins that can selectively bind to the particular unknown biopolymers.

12. A method of separating and identifying unknown proteins, the method comprising:

providing a nanopillar array having a substrate material that has a non-uniform gap size between nanopillars, the nanopillars having a coating of a different material from the substrate material that provides a uniform gap size, wherein the uniform gap size is about or below 100 nanometers;

introducing unknown proteins and protein-binding biopolymers into the nanopillar array, wherein when the unknown proteins bind with the protein-binding polymers, biopolymer-protein complexes are formed, and wherein when the unknown proteins do not bind with the protein-binding biopolymers, the unknown proteins and biopolymers remain unbound;

sorting the unknown proteins and the protein-binding biopolymers through the nanopillar array by transporting any of the unknown proteins and the protein-binding biopolymers which are unbound in a fluid flow direction, as the unknown proteins and the protein-binding biopolymers which are unbound are less than a predetermined bumping size;

sorting the unknown proteins and the protein-binding biopolymers through the nanopillar array by transporting the biopolymer-protein complexes in a bumped direction, as the biopolymer-protein complexes are at least the predetermined bumping size.
