Abstract

Microbiota restoration therapy compositions and methods for manufacturing, processing, and/or delivering microbiota restoration therapy compositions are disclosed. An example method for manufacturing a microbiota restoration therapy composition may include collecting a human fecal sample and adding a diluent to the human fecal sample to form a diluted sample. The diluent may include a cryoprotectant. The method may also include mixing the diluted sample with a mixing apparatus and filtering the diluted sample. Filtering may form a filtrate. The method may also include transferring the filtrate to a sample bag and sealing the sample bag.

Claims (52)

1. A method of treating a gastrointestinal disorder, the method comprising:

manufacturing a first microbiota restoration therapy composition, wherein manufacturing the first microbiota restoration therapy composition comprises:

collecting a human fecal sample;

adding a mixture of polyethylene glycol and saline having a concentration of 30-90 grams of polyethylene glycol per liter of saline to the human fecal sample to form a diluted sample;

mixing the diluted sample with a mixing apparatus;

filtering the diluted sample;

wherein filtering forms a filtrate;

transferring the filtrate to a sample bag; and

sealing the sample bag;

administering the first microbiota restoration therapy composition to a patient, the first microbiota restoration therapy composition derived from a first donor;

ascertaining a need for further treatment, wherein the need for further treatment comprises a reoccurrence of symptoms within 8 weeks of the administration of the first microbiota restoration therapy; and

administering a second microbiota restoration therapy composition to the patient when the need for further treatment occurs, the second microbiota restoration therapy composition derived from the first donor.

2. The method of claim 1 , wherein administering a first microbiota restoration therapy composition to a patient comprises administering one or more doses of the first microbiota restoration therapy composition.

3. The method of claim 1 , wherein administering a second microbiota restoration therapy composition to a patient comprises administering one or more doses of the second microbiota restoration therapy composition.

4. The method of claim 1 , further comprising administering a pre-treatment antibiotic.

5. The method of claim 1 , further comprising administering a post-treatment antibiotic.

6. The method of claim 1 , wherein administering a first microbiota restoration therapy composition to a patient comprises administering an enema.

7. The method of claim 1 , wherein administering a second microbiota restoration therapy composition to a patient comprises administering an enema.

8. The method of claim 1 , wherein the gastrointestinal disorder comprises a C. difficile infection.

9. A method of treating a gastrointestinal disorder, the method comprising:

manufacturing a microbiota restoration therapy composition and wherein manufacturing the microbiota restoration therapy composition comprises:

collecting a human fecal sample;

adding a mixture of polyethylene glycol and saline having a concentration of 30-90 grams of polyethylene glycol per liter of saline to the human fecal sample to form a diluted sample;

mixing the diluted sample with a mixing apparatus;

filtering the diluted sample;

wherein filtering forms a filtrate;

transferring the filtrate to a sample bag; and

sealing the sample bag;

administering a pre-treatment antibiotic to a patient with a gastrointestinal disorder;

administering a primary dosage of the microbiota restoration therapy composition to a patient;

ascertaining a need for further treatment, wherein the need for further treatment comprises a reoccurrence of symptoms within 8 weeks of the administration of the first microbiota restoration therapy; and

administering a secondary dosage of the same microbiota restoration therapy composition to the patient when the need for further treatment occurs.

10. The method of claim 9 , wherein the microbiota restoration therapy composition is derived from a single donor.

11. The method of claim 9 , wherein the pre-treatment antibiotic comprises vancomycin, fidaxomicin, rifaximimin, linezolid, daptomycin, quinupristin-dalfopristin, tigecycline, ceftaroline, ceftobiprole, televancin, teicoplanin, dalbavancin, metronidazole, ortivancin, or a combination thereof.

12. The method of claim 9 , further comprising administering a post-treatment antibiotic.

13. The method of claim 9 , wherein administering the first dosage of the microbiota restoration therapy composition to a patient comprises administering one or more individual doses of the microbiota restoration therapy composition.

14. The method of claim 9 , wherein administering the second dosage of the microbiota restoration therapy composition to a patient comprises administering one or more individual doses of the microbiota restoration therapy composition.

15. A method of treating a gastrointestinal disorder, the method comprising:

manufacturing the microbiota restoration therapy composition and wherein manufacturing the microbiota restoration therapy composition comprises:

collecting a human fecal sample;

adding a mixture of polyethylene glycol and saline having a concentration of 30-90 grams of polyethylene glycol per liter of saline to the human fecal sample to form a diluted sample;

mixing the diluted sample with a mixing apparatus;

filtering the diluted sample;

wherein filtering forms a filtrate;

transferring the filtrate to a sample bag; and

sealing the sample bag;

administering a pre-treatment antibiotic to a patient with a gastrointestinal disorder;

administering a primary dosage of a microbiota restoration therapy composition derived from a first donor to a patient, the microbiota restoration therapy composition pre-screened for bacterial diversity;

ascertaining a need for further treatment; and

administering a secondary dosage of the microbiota restoration therapy composition derived from the first donor to the patient.

16. The method of claim 15 , wherein the microbiota restoration therapy composition includes bacteria from at least seven different families and has a Shannon Diversity Index of 0.4-2.5 when calculated at the family level.

17. The method of claim 15 , wherein the primary dosage of a microbiota restoration therapy composition is administered with an enema tube.
