mRNA for use in treatment of human genetic diseases
View Patent ↗Compositions for modulating the expression of a protein in a target cell comprising at least one RNA molecule which comprises at least one modification 5 conferring stability to the RNA, as well as related methods, are disclosed.
1. A method of treating a urea cycle disorder comprising
administering to a subject suffering from or susceptible to a urea cycle disorder, a composition comprising an mRNA encoding an enzyme, encapsulated within a liposome;
wherein the enzyme is ornithine transcarbamylase (OTC), wherein the urea cycle disorder is OTC deficiency and
wherein the mRNA comprises a 5′ untranslated region (UTR) comprising SEQ ID NO: 1, wherein SEQ ID NO: 1 enhances the expression of the enzyme.
2. The method of claim 1 , wherein the mRNA is administered systemically.
3. The method of claim 1 , wherein the composition is administered intravenously.
4. The method of claim 1 , wherein the composition is administered bi-weekly.
5. The method of claim 1 , wherein the composition is administered monthly.
6. The method of claim 1 , wherein the administering of the composition results in increased expression of the enzyme encoded by the mRNA in the liver.
7. The method of claim 1 , wherein the mRNA comprises a modification to enhance stability.
8. The method of claim 7 , wherein the modification is selected from a group consisting of a modified nucleotide, an alteration to the 5′ or 3′ untranslated region (UTR), a cap structure and a poly A tail.
9. The method of claim 8 , wherein the modification comprises a pseudouridine.
