Methylphenidate-prodrugs, processes of making and using the same
View Patent ↗The present technology is directed to prodrugs and compositions for the treatment of various diseases and/or disorders comprising methylphenidate, or methylphenidate derivatives, conjugated to at least one alcohol, amine, oxoacid, thiol, or derivatives thereof. In some embodiments, the conjugates further include at least one linker. The present technology also relates to the synthesis of methylphenidate, or methylphenidate derivatives, conjugated to at least one alcohol, amine, oxoacid, thiol, or derivatives thereof or combinations thereof.
1. A composition comprising: at least one conjugate of methylphenidate, wherein the conjugate of methylphenidate has the following structure:
wherein G 2 is independently selected from the group consisting of standard amino acids, nonstandard amino acids and synthetic amino acids, wherein the amino acid is attached to the rest of the molecule by an amide linkage.
2. The composition of claim 1 , wherein G 2 is selected from the group consisting of Phe, His, Ile, and Pro.
3. The composition of claim 1 , wherein the conjugate of methylphenidate has the following structure:
4. The composition of claim 1 , wherein the conjugate of methylphenidate is Phe-(6-aminohexanoyl)-CH 2 OCO-MPH.
5. A composition comprising: at least one conjugate of methylphenidate, wherein the conjugate of methylphenidate has the following structure:
wherein G 2 and G 3 are independently selected from the group consisting of standard amino acids, nonstandard amino acids and synthetic amino acids, wherein the amino acid is attached to the rest of the molecule by an amide linkage.
6. The composition of claim 5 , wherein G 2 and G 3 are independently selected from the group consisting of Phe and Pro.
