Vectors and methods for genetic immunization
Improved DNA vaccine plasmids are disclosed that contain novel immunostimulatory RNA compositions. The improved plasmids eliminate all extraneous sequences, incorporate a novel antibiotic free short RNA based selectable marker, increase eukaryotic expression using a novel chimeric promoter, improve yield and stability during bacterial production, and improve immunostimulation. These vectors are utilized in immunization to elicit improved immune responses or therapy to induce type 1 interferon production.
1. A method for inducing an adaptive immune response in a mammal comprising the steps of:
a. providing a vector comprising a first RNA element comprising a VARNAI RNA component, and a second RNA element comprising an RIG-I activating immunostimulatory RNA element of 30-200 basepairs operably linked to a U6 RNA polymerase III promoter, and at least one gene encoding a protein or peptide antigen; and
b. delivering said vector to said mammal under conditions such that said antigen is expressed and said first RNA element and said second RNA element are expressed and induce an adaptive immune response to said antigen.
2. The method of claim 1 , wherein said vector is a non-viral vector.
3. The method of claim 1 , wherein said imunostimulatory RNA element of 30-200 basepairs is an U6 RNA Polymerase III promoter-expressed hairpin, double-stranded immunostimulatory RNA element.

The method of claim 1 , wherein said immunostimulatory RNA element of 30-200 base pairs operably linked to a U6 RNA polymerase III promoter further comprises a tRNA RNA polymerase promoter arranged in opposite orientation to said U6 RNA polymerase III promoter, such that transcription of said RNA element is convergent.
5. The method of claim 1 , whereby said adaptive immune response is increased, as compared to a vector lacking the combination of VARNAI and at least one additional immunostimulatory RNA element, without reducing antigen expression.
6. A method for inducing an adaptive immune response in a mammal comprising the steps of:
a. providing first and second vectors, said first vector comprising i) a first RNA element comprising a VARNAI RNA component, and ii) a gene encoding a protein or peptide antigen, said second vector comprising a second RNA element comprising an RIG-I activating immunostimulatory RNA element of 30-200 basepairs operably linked to a U6 RNA polymerase III promoter, and
b. delivering said first and second vectors to said mammal under conditions such that said antigen is expressed, and said first RNA element and said second RNA element are expressed and induce an adaptive immune response to said antigen.
7. The method of claim 6 , whereby said adaptive immune response is increased, as compared to vector (s) lacking the combination of VARNAI and at least one additional immunostimulatory RNA element, without reducing antigen expression.
8. A method for inducing type 1 interferon production in a mammal comprising the steps of:
a. obtaining a vector comprising a first RNA element comprising a VARNAI RNA component, and a second RNA element comprising an RIG-I activating immunostimulatory RNA element of 30-200 base pairs operably linked to a 136 RNA polymerase III promoter, wherein said immunostimulatory RNA element of 30-200 base pairs operably linked to a U6 RNA polymerase III promoter further comprises a tRNA RNA polymerase III promoter arranged in opposite orientation to said U6 RNA polymerase III promoter, such that transcription of said RNA element is convergent; and
b. delivering said vector to said mammal under conditions such that type 1 interferon production is induced.
9. The method of claim 8 , whereby said type 1 interferon production increases anti-viral, anti-tumor or antibacterial activity in said mammal.
10. The method of claim 8 , whereby said type 1 interferon production is increased, as compared to a vector lacking the combination of VARNAI and at least one additional immunostimulatory RNA element.
11. The method of claim 8 , wherein said imunostimulatory RNA element of 30-200 basepairs is an U6 RNA Polymerase III promoter-expressed hairpin, double-stranded immunostimulatory RNA element.
12. The method of claim 2 , wherein said non-viral vector is a plasmid vector.
