Conjugated anti-CD38 antibodies
View Patent ↗Isolated antibodies that bind to human CD38 and cynomolgus CD38 are disclosed. Also disclosed are pharmaceutical compositions comprising the disclosed antibodies, and therapeutic and diagnostic methods for using the disclosed antibodies.
1. An isolated antibody which specifically binds human CD38 protein (SEQ ID NO:1) and cynomolgus CD38 (SEQ ID NO:2), wherein said antibody interacts with at least K121, F135, Q139, D141, E239, W241, C275, K276, F284, P291 and E292 of SEQ ID NO:1 and SEQ ID NO:2, based on human sequence numbering, and
wherein said antibody is covalently attached to a drug moiety.
2. An isolated antibody according to claim 1 , wherein said drug moiety is selected from the group consisting of an auristatin, a maytansinoid, and a calicheamicin, a dolstatin, and a trichothecene.
3. An isolated antibody according claim 1 , wherein said drug moiety is covalently attached to said antibody by a linker.
4. An isolated antibody according claim 3 , wherein said linker is cleavable.
5. An isolated antibody according claim 4 , wherein said linker is cleavable under intracellular or extracellular conditions.
6. An isolated antibody according to claim 1 , further comprising an Fc domain.
7. An isolated antibody according to claim 6 , wherein said Fc domain is human.
8. An isolated antibody according to claim 6 , wherein said Fc domain is a variant Fc domain.
9. An isolated nucleic acid encoding the heavy chain of the antibody of claim 1 .
10. An isolated nucleic acid encoding the light chain of the antibody of claim 1 .
11. A host cell comprising the isolated nucleic acids of claim 9 or 10 .

A method of producing an isolated antibody which specifically binds human CD38 protein (SEQ ID NO:1) and cynomolgus CD38 (SEQ ID NO:2), wherein said antibody interacts with at least K121, F135, Q139, D141, E239, W241, C275, K276, F284, P291 and E292 of SEQ ID NO:1 and SEQ ID NO:2, based on human sequence numbering, and wherein said antibody is covalently attached to a drug moiety, comprising culturing the host cell of claim 11 under conditions wherein said antibody is produced.

A method of treating a cancer expressing CD38 comprising administering to a patient in need thereof an isolated antibody which specifically binds human CD38 (SEQ ID NO:1) and cynomolgus CD38 (SEQ ID NO:2), wherein said antibody interacts with at least K121, F135, Q139, D141, E239, W241, C275, K276, F284, P291 and E292 of SEQ ID NO:1 and SEQ ID NO:2, based on human sequence numbering, and
wherein said antibody is covalently attached to a drug moiety.
14. A method of treating according to claim 13 , wherein said drug moiety is selected from the group consisting of an auristatin, a maytansinoid, and a calicheamicin, a dolstatin, and a trichothecene.
15. A method of treating according claim 13 , wherein said drug moiety is covalently attached to said antibody by a linker.
16. A method of treating according claim 15 , wherein said linker is cleavable.
17. A method of treating according claim 16 , wherein said linker is cleavable under intracellular or extracellular conditions.
18. A method of treating according to claim 13 , further comprising an Fc domain.
19. A method of treating according to claim 18 , wherein said Fc domain is human.
20. A method of treating according to claim 18 , wherein said Fc domain is a variant Fc domain.
