Abstract

Compositions comprising hydrocodone benzoic acid enol ester to form novel prodrugs including hydrocodone benzoic acid enol ester salts, and various polymorphs. Also provided are processes for the preparation of hydrocodone benzoic acid enol ester salts, and various polymorphs.

Claims (31)

1. A compound comprising a polymorphic form of hydrocodone benzoic acid enol ester hydrochloride, wherein the polymorphic form is Form II, Form III, Form IV, Form V, or a combination thereof wherein:

a. Form II has an XRPD pattern comprising peaks at about the following 2θ values: 4.31, 8.62, and 15.72;

c. Form III has an XRPD pattern comprising peaks at about the following 2θ values: 4.35, 8.68, 13.01, and 15.05;

c. Form IV has an XRPD pattern comprising peaks at about the following 2θ values: 7.87, 10.27, 13.18, 14.19, 14.65 and 18.58; and

d. Form V has an XRPD pattern comprising peaks at about the following 2θ values: 7.50, 9.04, 13.13, and 21.10.

2. The compound of claim 1 , wherein:

a. Form II has an x-ray powder diffraction pattern (XRPD) further comprising peaks at about the following 2θ values: 12.95, 13.19, 13.42, 14.33, 14.97, 17.48, 18.47, 21.73, 22.92, 24.15, 24.98, 26.41, 26.53, 26.62, 27.93, 30.46;

b. Form III has an XRPD pattern further comprising peaks at about the following 2θ values: 13.27, 13.86, 14.06, 16.02, 18.40, 21.72, 22.38, 26.14;

c. Form IV has an XRPD pattern further comprising peaks at about the following 2θ values: 11.82, 12.56, 15.34, 16.77, 17.67, 19.50, 20.30, and 21.31; and

d. Form V has an XRPD pattern further comprising peaks at about the following 2θ values: 6.17, 10.96, 11.93, 12.34, 12.64, 14.34, 14.62, 14.93, 16.73, 17.04, 17.22, 17.81, 18.59, 18.98, 19.19, 19.52, 20.67, 21.91, 22.15, 22.82, 23.49, 25.03, 25.37, 26.09, and 26.43.

A solid pharmaceutical composition comprising a therapeutically effective amount of a polymorphic form of hydrocodone benzoic acid enol ester hydrochloride and a pharmaceutically acceptable excipient in a formulation for oral administration, wherein the polymorphic form is Form II, III, IV, V, or a combination thereof wherein:

a. Form II has an XRPD pattern comprising peaks at about the following 20 values: 4.31, 8.62, and 15.72;

b. Form III has an XRPD pattern comprising peaks at about the following 20 values: 4.35, 8.68, 13.01, and 15.05;

c. Form IV has an XRPD pattern comprising peaks at about the following 20 values: 7.87, 10.27, 13.18, 14.19, 14.65 and 18.58; and

d. Form V has an XRPD pattern comprising peaks at about the following 20 values: 7.50, 9.04, 13.13, and 21.10.

4. The solid pharmaceutical composition of claim 3 , wherein:

a. Form II has an x-ray powder diffraction pattern (XRPD) pattern further comprising peaks at about the following 2θ values: 12.95, 13.19, 13.42, 14.33, 14.97, 17.48, 18.47, 21.73, 22.92, 24.15, 24.98, 26.41, 26.53, 26.62, 27.93, 30.46;

b. Form III has an XRPD pattern further comprising peaks at about the following 2θ values: 13.27, 13.86, 14.06, 16.02, 18.40, 21.72, 22.38, 26.14;

c. Form IV has an XRPD pattern further comprising peaks at about the following 2θ values: 11.82, 12.56, 15.34, 16.77, 17.67, 19.50, 20.30, and 21.31; and

d. Form V has an XRPD pattern further comprising peaks at about the following 2θ values: 6.17, 10.96, 11.93, 12.34, 12.64, 14.34, 14.62, 14.93, 16.73, 17.04, 17.22, 17.81, 18.59, 18.98, 19.19, 19.52, 20.67, 21.91, 22.15, 22.82, 23.49, 25.03, 25.37, 26.09, and 26.43.

5. A process for preparing the polymorphic form II of hydrocodone benzoic acid enol ester hydrochloride of claim 1 , the process comprising recrystallizing hydrocodone benzoic acid enol ester hydrochloride from an organic solvent containing less than 0.2% water.

6. The process of claim 5 , wherein substantially pure polymorphic Form II is formed by the process.

7. The process of claim 5 , wherein the recrystallizing step comprises:

a. adding anhydrous isopropanol and isopropyl acetate to solid hydrocodone benzoic acid enol ester hydrochloride to create a slurry;

b. heating the slurry to reflux to create a solution;

c. cooling the solution to create substantially pure crystalline Form II.

8. The process of claim 6 , further comprising:

heating the polymorphic Form II at a temperature of between about 210° C. and about 230° C. to form polymorphic Form III.

9. The process of claim 6 , further comprising: exposing polymorphic Form II to a relative humidity of from about 75% to about 100% for a period of time to form polymorphic Form IV.

10. The process of claim 8 , further comprising: exposing the polymorph Form III to a relative humidity from about 75% to about 100% for a period of time to form polymorphic Form V.

11. A method of treating pain, the method comprising administering to a human in need thereof, a pharmaceutical composition comprising a therapeutically effective amount of one or more polymorphic forms of hydrocodone benzoic acid enol ester hydrochloride the compound of claim 1 .
