Abstract

The present application provides methods for preparing soluble lipidated ligand agents comprising a ligand entity and a lipid entity, and in some embodiments, provides relevant parameters of each of these components, thereby enabling appropriate selection of components to assemble active agents for any given target of interest.

Claims (20)

1. A soluble lipidated ligand agent comprising:

a ligand entity comprising:

(a) a human chemerin fragment having the sequence of YFPGQFAFS (SEQ ID NO.: 8) or

(b) a chemerin analog having the sequence of YFLPSQFA-Tic-S(SEQ ID NO.: 10), wherein one or more of the amino acids are D-amino acids, and wherein Tic represents (S)-1,2,3,4-tetrahydroisoquinoline-3-carboxylic acid;

at least one lipid entity covalently linked to the ligand entity at or near the N-terminus, at or near the C-terminus, or at or near both the N-terminus and C-terminus; and

an optional linker entity connecting the ligand entity to the at least one lipid entity.

2. The soluble lipidated ligand agent of claim 1 , wherein the ligand entity binds to a membrane-associated target, a cell-surface-associated target, or an intracellular target.

3. The soluble lipidated ligand agent of claim 2 , wherein the membrane-associated target is selected from the group consisting of a receptor, an enzyme, and an ion channel.

4. The soluble lipidated ligand agent of claim 1 , wherein the linker entity is formed at least in part as a result of a CLICK reaction.

5. The soluble lipidated ligand agent of claim 4 , wherein the CLICK reaction is an azide-alkyne Huisgen cycloaddition reaction.

6. The soluble lipidated ligand agent of claim 1 , wherein the linker comprises at least one molecule of polyethylene glycol (PEG).

7. The soluble lipidated ligand agent of claim 5 , wherein the lipid entity is selected from the lipid entities listed in Table 3.

8. A composition comprising:

a soluble lipidated ligand agent according to claim 1 ; and

a pharmaceutically acceptable carrier.

9. The soluble lipidated ligand agent of claim 1 , wherein the lipid entity is palmitic acid.

10. The soluble lipidated ligand agent of claim 1 , wherein the Y, S, and A in YFLPSQFA-Tic-S(SEQ ID NO.: 10) are D-amino acids.

11. The soluble lipidated ligand agent of claim 3 , wherein the receptor is CMKLR1.

12. A soluble lipidated ligand agent having the following structure:

wherein Tic represents (S)-1,2,3,4-tetrahydroisoquinoline-3-carboxylic acid, and wherein the GGYFLPSQFA-Tic-S is SEQ ID NO.: 11.
