Abstract

The present invention is directed compositions for targeted delivery of RNA interference (RNAi) polynucleotides to hepatocytes in vivo. Targeted RNAi polynucleotides are administered together with co-targeted melittin delivery peptides. Delivery peptides provide membrane penetration function for movement of the RNAi polynucleotides from outside the cell to inside the cell. Reversible modification provides physiological responsiveness to the delivery peptides.

Claims (20)

1. A peptide comprising the amino acid sequence of Seq ID 2, Seq ID 3, Seq ID 4, Seq ID 5, Seq ID 6, Seq ID 7, Seq ID 8, Seq ID 9, Seq ID 10, or Seq ID 11.

2. The peptide of claim 1 wherein the peptide consists of L-form amino acids.

3. The peptide of claim 1 wherein the Melittin peptide contains at least one D-form amino acid.

4. The peptide of claim 1 wherein at least one targeting ligand is linked to an amine on the peptide via a physiologically labile reversible linkage.

5. The peptide of claim 4 wherein the physiologically labile reversible linkage is a disubstituted maleamate.

6. The peptide of claim 5 wherein the targeting ligand comprises an Asialoglycoprotein Receptor (ASGPr) ligand.

7. The peptide of claim 6 wherein the ASGPr ligand is selected from the group consisting of lactose, galactose, N-acetylgalactosamine (GalNAc), galactosamine, N-formylgalactosamine, N-propionylgalactosamine, N-n-butanoyl-galactosamine, and N-iso-butanoyl-galactosamine.

8. The peptide of claim 1 wherein a polyethyleneglycol (PEG) is covalently linked to the amino terminus of the peptide.

9. The peptide of claim 8 wherein at least one targeting ligand is linked to a lysine amine on the peptide via a physiologically labile reversible linkage.

10. The peptide of claim 9 wherein the physiologically labile reversible linkage is a disubstituted maleamate.

11. The peptide of claim 10 wherein the targeting ligand comprises an ASGPr ligand.

12. The peptide of claim 11 wherein the ASGPr ligand is selected from the group consisting of lactose, galactose, GalNAc, galactosamine, N-formylgalactosamine, N-propionylgalactosamine, N-n-butanoyl-galactosamine, and N-iso-butanoyl-galactosamine.

13. The peptide of claim 1 wherein a first targeting ligand-PEG conjugate is covalently linked to the amino terminus of the peptide.

14. The peptide of claim 13 wherein the first targeting ligand comprises a first ASGPr ligand.

15. The peptide of claim 14 wherein the first ASGPr ligand is selected from the group consisting of lactose, galactose, GalNAc, galactosamine, N-formylgalactosamine, N-propionylgalactosamine, N-n-butanoyl-galactosamine, and N-iso-butanoyl-galactosamine.

16. The peptide of claim 15 wherein at least one second targeting ligand is linked to a lysine amine on the peptide via a physiologically labile reversible linkage.

17. The peptide of claim 16 wherein the physiologically labile reversible linkage is a disubstituted maleamate.

18. The peptide of claim 17 wherein the second targeting ligand comprises an ASGPr ligand.

19. The peptide of claim 17 wherein the second ASGPr ligand is selected from the group consisting of lactose, galactose, GalNAc, galactosamine, N-formylgalactosamine, N-propionylgalactosamine, N-n-butanoyl-galactosamine, and N-iso-butanoyl-galactosamine.

20. The peptide of claim 7 wherein the peptide is Seq ID 7 and the ASGPr ligand is GalNAc.
