Abstract

A depth filtration device for the clarification of biological fluids including a composite depth filter media having a nonwoven first layer integral with a second layer containing a polyaeryionitrile (PAN) fibers, a filter aid, and a wet-strength resin. The depth filter media exhibits increased creased binding capacity for soluble impurities such as DNA and host eel! proteins from biological/cell culture feed-streams during secondary clarification and low-level impurity clearance of harvested cell culture fluids, such as those used for the manufacture of monoclonal antibodies. The depth filter media additionally exhibits significantly lower flushing requirements, resulting in lower levels of organic, inorganic and bioburden extractables released, high dirt holding capacities and good chemical and/or radiation resistance.

Claims (5)

1. A synthetic composite depth filter media consisting of a mixture of fibrillated polyacrylonitrile fibers consisting of polyacrylonitrile or polyacrylonitrile copolymers and a silica filter aid consisting of silica particles, wherein the silica filter aid consists of fine silica particles less than or equal to about 5 μm and coarse silica particles greater than 5 μm and less than or equal to about 40 μm, and a charged wet strength resin.

2. The synthetic composite depth filter media according to claim 1 , wherein the filter aid is over 0 to 90% by weight based on the total weight of the fibrillated polyacrylonitrile fibers and filter aid.

The synthetic composite depth filter media according to claim 1 , wherein the wet strength resin consists of one or more of water-soluble synthetic polymers of urea or melamine-formaldehyde based polymers, polyaminopolyamide-epichlorohydrin (PAE) polymers or glyoxalated polyacrylamide (GPAM) resins.

4. The synthetic composite depth filter media according to claim 3 , wherein the content by weight of the wet strength resin based on the total weight of the fibrillated polyacrylonitrile fibers and filter aid is over 0% to about 5%.

5. The synthetic composite depth filter media of claim 1 , wherein the content by weight of the fibrillated polyacrylonitrile fibers are about 25% to about 50% by weight of the fibrillated polyacrylonitrile fibers and filter aid.
