Abstract

The present disclosure provides immunogenic compositions useful in prevention and treatment of Staphylococcus aureus infection. In particular, the disclosure provides delta toxin and phenol-soluble modulin peptides as well as mutants, fragments, variants or derivatives thereof. The disclosure further provides multivalent oligopeptides, fusion proteins comprising two or more staphylococcal proteins, e.g., DT, PSM, alpha hemolysin, leukocidin, superantigen, or any fragments, variants, derivatives, or mutants thereof fused together as a single polypeptide in any order.

Claims (12)

1. A multivalent oligopeptide, comprising a fusion of three or more Staphylococcus aureus -derived peptides, or mutants, fragments, variants, or derivatives thereof arranged in any order, wherein the multivalent oligopeptide comprises three or more superantigen (SAg) polypeptides, or mutants, fragments, variants, or derivatives thereof, and wherein three of the three or more SAg polypeptides, or mutants, fragments, variants, or derivatives thereof comprise SEQ ID NO: 49, SEQ ID NO: 50, and SEQ ID NO: 51.

2. The oligopeptide of claim 1 , wherein the three or more Staphylococcus aureus -derived peptides, or mutants, fragments, variants, or derivatives thereof are associated via linkers.

3. The oligopeptide of claim 1 , comprising the amino acid sequence SEQ ID NO: 32, SEQ ID NO: 33, SEQ ID NO. 34, SEQ ID NO: 35, SEQ ID NO: 36, SEQ ID NO: 37, or any combination thereof.

4. The oligopeptide of claim 1 , further comprising a heterologous peptide or polypeptide.

5. The oligopeptide of claim 1 , further comprising an immunogenic carbohydrate.

6. A composition comprising the oligopeptide of claim 1 , and a carrier.

7. The composition of claim 6 , further comprising an adjuvant.

8. The composition of claim 6 , further comprising an immunogen.

9. A method of inducing a host immune response against Staphylococcus aureus , comprising administering to a subject in need of the immune response an effective amount of the composition of claim 6 .

10. A method of treating a Staphylococcal disease or infection in a subject comprising administering to a subject in need thereof the composition of claim 6 .

11. A method of producing a vaccine against S. aureus infection comprising:

isolating the oligopeptide of claim 1 and combining the oligopeptide with an adjuvant.
