Diagnostic assays and kits for detection of folate receptor 1
The invention generally relates to antibodies that bind to human folate receptor 1 and diagnostic assays for folate receptor 1-based therapies. Methods of using the antibodies to monitor therapy are further provided.
1. A method of detecting FOLR1 protein in a sample comprising contacting said sample with an antibody or antigen-binding fragment thereof wherein the antibody or antigen-binding fragment binds to the same FOLR1 epitope as an antibody selected from the group consisting of:
(a) an antibody comprising the variable heavy chain polypeptide of SEQ ID NO: 25 and the variable light chain polypeptide of SEQ ID NO: 29; and
(b) an antibody comprising the variable heavy chain polypeptide of SEQ ID NO: 26 and the variable light chain polypeptide of SEQ ID NO: 30.
2. The method of claim 1 , wherein said sample is a cancer sample and said cancer is selected from the group consisting of: ovarian, non-small cell lung cancer, uterine, endometrial, pancreatic, renal, lung, cancer of the peritoneum, and breast cancer.
3. The method of claim 1 , wherein said antibody or antigen-binding fragment thereof used to detect FOLR1 protein is a murine, chimeric, humanized, or resurfaced antibody or antigen-binding fragment thereof.
4. The method of claim 1 , wherein said antibody or antigen-binding fragment thereof used to detect FOLR1 protein binds to a human folate receptor 1 with a Kd of about 1.0 nM to about 10 nM.
5. The method of claim 1 , wherein the antibody or antigen-binding fragment thereof used to detect FOLR1 protein comprises a detection agent.
6. The method of claim 5 , wherein the detection agent is a chromogenic detection agent, a fluorogenic detection agent, an immunofluorescent detection agent, an enzymatic detection agent, or an electr chemiluminescent detection agent.
7. The method of claim 1 , wherein the antibody or antigen-binding fragment binds to the same FOLR1 epitope as an antibody comprising the variable heavy chain polypeptide of SEQ ID NO: 25 and the variable light chain polypeptide of SEQ ID NO: 29.
8. The method of claim 1 , wherein the antibody or antigen-binding fragment binds to the same FOLR1 epitope as an antibody comprising the variable heavy chain polypeptide of SEQ NO: 26 and the variable light chain polypeptide of SEQ ID NO: 30.
9. The method of claim 1 , wherein the FOLR1 protein is detected using an antibody or antigen-binding fragment thereof, which comprises variable heavy chain CDR1, CDR2, and CDR3 and variable light chain CDR1, CDR2, and CDR3 amino acid sequences selected from the group consisting of:
(a) SEQ ID NOs: 1, 2, and 3 and SEQ ID NOs: 13, 14, and 15, respectively; and
(b) SEQ ID NOs: 4, 5, and 6 and SEQ ID NOs: 16, 17, and 18, respectively.
10. The method of claim 9 , wherein the FOLR1 protein is detected using an antibody or antigen-binding fragment thereof that comprises variable heavy chain CDR1, CDR2, and CDR3 and variable light chain CDR1, CDR2, and CDR3 amino acid sequences comprising SEQ ID NOs: 4, 5, and 6 and SEQ ID NOs: 16, 17, and 18, respectively.
11. The method of claim 9 , wherein the FOLR1 protein is detected using an antibody or antigen-binding fragment thereof that comprises the variable heavy chain polypeptide of SEQ ID NO: 25 and the variable light chain polypeptide of SEQ ID NO: 29.
12. The method of claim 9 , wherein FOLR1 protein is detected using an antibody or antigen-binding fragment thereof that comprises the variable heavy chain polypeptide of SEQ ID NO: 26 and the variable light chain polypeptide of SEQ ID NO: 30.
13. The method of claim 9 , wherein the sample is a cancer sample.
14. The method of claim 9 , wherein the FOLR1 protein is detected using an antibody or antigen-binding fragment thereof that comprises variable heavy chain CDR1, CDR2, and CDR3 and variable light chain CDR1, CDR2, and CDR3 amino acid sequences comprising SEQ ID NOs: 1, 2, and 3 and SEQ ID NOs: 13, 14, and 15, respectively.
15. The method of claim 9 , wherein the FOLR1 protein is detected using an antibody that comprises the heavy chain amino acid sequence of SEQ ID NO: 33 and the light chain amino acid sequence of SEQ ID NO: 37.
16. The method of claim 9 , wherein the FOLR1 protein is detected using an antibody that comprises the heavy chain amino acid sequence of SEQ ID NO: 34 and the light chain amino acid sequence of SEQ ID NO: 38.
17. The method of claim 9 , wherein the FOLR1 protein is shed FOLR1 protein.
18. The method of claim 9 , wherein the sample is a bodily fluid sample.
19. The method of claim 13 , further comprising administering to a patient with a FOLR1-positive cancer an active agent comprising a FOLR1 antibody or antigen-binding fragment thereof with the variable heavy chain sequence of SEQ ID NO: 46 and the variable light chain sequence of SEQ ID NO: 48.
20. The method of claim 13 , wherein the FOLR1 protein is shed FOLR1 protein.
21. The method of claim 13 , wherein the sample is a bodily fluid sample.
22. The method of claim 19 , wherein the active agent comprises a FOLR1 antibody comprising a heavy chain comprising the same amino acid sequence as the amino acid sequence of the heavy chain encoded by the plasmid deposited with the American Type Culture Collection (ATCC) as PTA-10772 and a light chain comprising the same amino acid sequence as the amino acid sequence of the light chain encoded by the plasmid deposited with the ATCC as PTA-10774.
23. The method of claim 19 , wherein the active agent comprises the FOLR1 antibody or antigen-binding fragment thereof conjugated to a cytotoxic agent.
24. The method of claim 22 , wherein the active agent comprises the FOLR1 antibody or antigen-binding fragment thereof conjugated to a cytotoxic agent.
25. The method of claim 23 , wherein the active agent comprises the FOLR1 antibody or antigen-binding fragment thereof conjugated to the maytansinoid DM4 via the cleavable sulfo-SPDB linker.
26. The method of claim 24 , wherein the active agent comprises the FOLR1 antibody or antigen-binding fragment thereof conjugated to the maytansinoid DM4 via the cleavable sulfo-SPDB linker.
27. The method of claim 26 , wherein the cancer is ovarian cancer.
