Abstract

The present invention provides antigen-binding proteins capable of binding to KIR3D polypeptides. The antibodies have increased activity in the treatment of disorders characterized by KIR3DL2-expressing cells, particularly CD4+ T cells, including malignancies such as Mycosis Fungoides and Sézary Syndrome.

Claims (7)

1. A method of modulating T cell activity in a patient suffering from arthritis, rheumatoid arthritis, or spondylarthritis comprising the step of administering to said patient a composition comprising a pharmaceutically acceptable carrier or excipient and an antibody that specifically binds to an epitope within the KIR3DL2 domain 0 consisting of SEQ ID NO: 21, wherein said antibody inhibits proliferation of KIR3DL-expressing human T lymphocytes and comprises a constant region of human IgG isotype, wherein said antibody: (a) comprises an Fc region of an IgG isotype modified to decrease binding to Fc receptors; and/or (b) does not substantially induce ADCC of a KIR3DL-expressing target cell.

2. The method of claim 1 , further comprising administering to the patient an immune system suppressor.

3. The method of claim 1 , wherein said patient suffers from arthritis.

4. The method of claim 1 , wherein said patient suffers from rheumatoid arthritis.

5. The method of claim 1 , wherein said patient suffers from spondylarthritis.

6. The method of claim 1 , wherein said antibody does not substantially induce ADCC of a KIR3DL-expressing target cell.

7. The method of claim 1 , wherein said antibody comprises an Fc region of an IgG isotype modified to decrease binding to Fc receptors.
