View Patent ↗Disclosed are novel compounds that are useful in regulating the expression of interleukin-6 (IL-6) and/or vascular cell adhesion molecule-1 (VCAM-1), and their use in the treatment and/or prevention of cardiovascular and inflammatory diseases and related disease states, such as, for example, atherosclerosis, asthma, arthritis, cancer, multiple sclerosis, psoriasis, and inflammatory bowel diseases, and autoimmune disease(s). Also, disclosed are compositions comprising the novel compounds, as well as methods for their preparation.
1. A method for reducing IL-6 and/or VCAM-1 in a subject, comprising administering a therapeutically effective amount of at least one compound of Formula I:
or a stereoisomer, tautomer, pharmaceutically acceptable salt, or hydrate thereof, wherein:
Q and V are independently selected from CH and nitrogen;
U is selected from C═O and SO 2 ;
Rc is selected from hydrogen, C 1 -C 6 alkyl, and C 3 -C 6 cycloalkyl;
Ra 1 , Ra 2 , and Ra 3 are independently selected from hydrogen, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, C 1 -C 6 alkoxy, halogen, amino, amide, hydroxyl, heterocycle, and C 3 -C 6 cycloalkyl, wherein Ra 1 and Ra 2 and/or Ra 2 and Ra 3 may be connected to form a cycloalkyl or a heterocycle;
Rb 2 and Rb 6 are independently selected from hydrogen, halogen, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 3 -C 6 cycloalkyl, hydroxyl, and amino;
Rb 3 and Rb 5 are independently selected from hydrogen, halogen, C 1 -C 6 alkyl, C 1 -C 6 alkoxy, C 3 -C 6 cycloalkyl, hydroxyl, and amino, wherein
Rb 2 and Rb 3 and/or Rb 5 and Rb 6 may be connected to form a cycloalkyl or a heterocycle;
represents a 3-8 membered ring system wherein:
W is selected from carbon and nitrogen;
Z is selected from CR 6 R 7 , NR 8 , oxygen, sulfur, —S(O)—, and —SO 2 —;
said ring system being optionally fused to another ring selected from cycloalkyl, heterocycle, and phenyl, and wherein said ring system is optionally selected from rings having the structures
R 3 , R 4 , and R 5 are independently selected from hydrogen, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, C 1 -C 6 alkoxy, C 3 -C 6 cycloalkyl, aryl, aryloxy, hydroxyl, amino, amide, oxo, —CN, and sulfonamide;
R 6 and R 7 are independently selected from hydrogen, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, C 3 -C 6 cycloalkyl, aryl, halogen, hydroxyl, acyl, and —CN; and
R 8 is selected from hydrogen, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, acyl, and C 3 -C 6 cycloalkyl; and
R 9 , R 10 , R 11 , and R 12 are independently selected from hydrogen, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, C 3 -C 6 cycloalkyl, aryl, hydroxyl, sulfonyl, and acyl,
provided that
if Q=CH, then at least one of Ra 1 , Ra 2 , and Ra 3 is not hydrogen;
if Z═NAc, then only one of Ra 1 , Ra 2 , and Ra 3 is hydrogen, and Ra 1 is not —OCH 2 CH 2 OMe;
if Ra 1 and Ra 3 are both OMe, then R 8 is not —C(O)CH 2 OH; and
further provided that the compound of Formula I is not 5,7-dimethoxy-2-(4-morpholinophenyl)quinazolin-4(3H)-one, 5,7-dimethoxy-2-(4-(4-methylpiperazin-1-yl)phenyl)quinazolin-4(3H)-one, or 2-(4-(1-cyclopentylpiperidin-4-yl)phenyl)-3-methylquinazolin-4(3H)-one.
2. The method according to claim 1 , wherein
R 3 and R 4 are independently selected from hydrogen, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, C 1 -C 6 alkoxy, C 3 -C 6 cycloalkyl, aryloxy, aryl, hydroxyl, amino, amide, oxo, —CN, and sulfonamide; and
R 8 is selected from hydrogen, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, acyl, C 3 -C 8 cycloalkyl, and C 2 -C 6 alkynyl.
3. The method according to claim 1 , wherein
R 3 and R 4 are independently selected from hydrogen, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, C 1 -C 6 alkoxy, C 3 -C 6 cycloalkyl, aryloxy, aryl, hydroxyl, amino, amide, oxo, —CN, and sulfonamide; and
R 9 and R 10 are independently selected from hydrogen, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, C 3 -C 6 cycloalkyl, aryl, heterocycle, sulfonyl, carbamate, carboxamide, and acyl.
4. The method according to claim 1 , wherein
R 3 and R 4 are independently selected from hydrogen, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, C 1 -C 6 alkoxy, C 3 -C 6 cycloalkyl, aryloxy, aryl, hydroxyl, amino, amido, oxo, —CN, and sulfonamide; and
R 8 is selected from hydrogen, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, acyl, and C 3 -C 6 cycloalkyl.
5. The method according to claim 1 , wherein
U is C═O;
Rc is hydrogen;
Ra 2 is hydrogen;
Ra 1 and Ra 3 are independently selected from C 1 -C 6 alkoxy, hydrogen, and halogen;
Rb 2 , Rb 3 , Rb 5 , and Rb 6 are each hydrogen;
is selected from
R 3 and R 4 are independently selected from hydrogen and C 1 -C 6 alkyl;
R 5 is selected from C 1 -C 6 alkyl and hydrogen; and
R 9 , R 10 , R 11 , and R 12 are independently selected from C 1 -C 6 alkyl, hydrogen, acyl, and sulfonyl.
6. The method according to claim 1 , wherein
U is C═O;
Rc is hydrogen;
Ra 2 is hydrogen;
Ra 1 and Ra 3 are independently selected from methoxy, hydrogen, and halogen;
Rb 2 , Rb 3 , Rb 5 , and Rb 6 are each hydrogen;

7. A method for reducing IL-6 and/or VCAM-1 in a subject, comprising administering a therapeutically effective amount of at least one compound selected from:

8. The method according to claim 1 , wherein the therapeutically effective amount of the compound is administered with at least one pharmaceutically acceptable carrier in a pharmaceutically acceptable composition.
9. The method according to claim 1 , further comprising treating and/or reducing the risk of acquiring cardiovascular disorders, inflammatory diseases, and/or cancer, characterized by altered expression of IL-6 and/or VCAM-1 proliferation.
10. The method according to claim 9 , wherein the subject is a human.
11. A compound of Formula I:

further provided that the compound of Formula I is not 5,7-dimethoxy-2-(4-morpholinophenyl)quinazolin-4(3H)-one, 5,7-dimethoxy-2-(4-(4-methylpiperazin-1-yl)phenyl)quinazolin-4(3H)-one, or 2-(4-(1-cyclopentylpiperidin-4-yl)phenyl)-3-methylquinazolin-4(3H)-one.
12. The compound according to claim 11 , wherein
R 3 and R 4 are independently selected from hydrogen, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, C 1 -C 6 alkoxy, C 3 -C 6 cycloalkyl, aryloxy, aryl, hydroxyl, amino, amide, oxo, —CN, and sulfonamide; and
R 8 is selected from hydrogen, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, acyl, and C 3 -C 6 cycloalkyl.
13. The compound according to claim 11 , wherein
R 3 and R 4 are independently selected from hydrogen, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, C 1 -C 6 alkoxy, C 3 -C 6 cycloalkyl, aryloxy, aryl, hydroxyl, amino, amide, oxo, —CN, and sulfonamide; and
R 9 and R 10 are independently selected from hydrogen, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, C 3 -C 6 cycloalkyl, aryl, heterocycle, sulfonyl, carbamate, carboxamide, and acyl.
14. The compound according to claim 11 , wherein
R 3 and R 4 are independently selected from hydrogen, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, C 1 -C 6 alkoxy, C 3 -C 6 cycloalkyl, aryloxy, aryl, hydroxyl, amino, amide, oxo, —CN, and sulfonamide; and
R 9 and R 10 are independently selected from hydrogen, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, C 3 -C 6 cycloalkyl, aryl, heterocycle, sulfonyl, carbamate, carboxamide, and acyl.
15. The compound according to claim 11 , wherein
R 3 and R 4 are independently selected from hydrogen, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, C 1 -C 6 alkoxy, C 3 -C 6 cycloalkyl, aryloxy, aryl, hydroxyl, amino, amide, oxo, —CN, and sulfonamide; and
R 8 is selected from hydrogen, C 1 -C 6 alkyl, C 2 -C 6 alkenyl, C 2 -C 6 alkynyl, acyl, and C 3 -C 6 cycloalkyl.
16. The compound according to claim 11 , wherein

20. A method for reducing IL-6 and/or VCAM-1 in a subject, comprising administering a composition according to claim 19 .
21. The method according to claim 20 , further comprising treating and/or reducing the risk of acquiring cardiovascular disorders, and inflammatory diseases, and/or cancer, characterized by altered expression of IL-6 and/or VCAM-1 proliferation.
22. The method according to claim 21 , wherein the subject is a human.

The method according to claim 9 , wherein the inflammatory disease is selected from arthritis, rheumatoid arthritis, asthma, dermatitis, psoriasis, cystic fibrosis, post transplantation late and chronic solid organ rejection, multiple sclerosis, systemic lupus erythematosus, inflammatory bowel diseases, autoimmune diabetes, diabetic retinopathy, diabetic nephropathy, diabetic vasculopathy, ocular inflammation, uveitis, rhinitis, ischemic-reperfusion injury, post-angioplasty restenosis, chronic obstructive pulmonary disease (COPD), glomerulonephritis, Grave's disease, gastrointestinal allergies, and conjunctivitis.

The method according to claim 9 , wherein the cancer is selected from multiple myeloma, lymphoma, leukemia, solid tumors, prostate and bladder cancers, cardiac myxoma, cerebral edema secondary to brain tumors, hormone-independent prostate cancer, B cell lymphoma, AIDS-associated lymphoma, and metastatic renal cell carcinoma.

27. A pharmaceutical composition comprising a compound of claim 7 and a pharmaceutically acceptable carrier.
28. A method for treating and/or reducing, the risk of acquiring cardiovascular disorders, inflammatory diseases, and/or cancer, characterized by altered expression of IL-6 and/or VCAM-1 proliferation, comprising administering a composition according to claim 27 .
29. The method according to claim 28 , wherein the subject is a human.

32. A pharmaceutical composition comprising a compound of claim 18 and a pharmaceutically acceptable carrier.
33. A method for reducing IL-6 and/or VCAM-1 in a subject, comprising administering a composition according to claim 32 .
34. The method according to claim 33 , further comprising treating and/or reducing the risk of acquiring cardiovascular disorders, inflammatory diseases, and/or cancer, characterized by altered expression of IL-6 and/or VCAM-1 proliferation.
35. The method according to claim 34 , wherein the subject is a human.

38. The method according to claim 7 , wherein the compound is selected from: 5,7-dimethoxy-2-(4-(piperazin-1-yl)phenyl)quinazolin-4(3H)-one; 2-(4-(4-acetyl-3-methylpiperazin-yl)phenyl)-5,7-dimethoxyquinazolin-4(3H)-one; and pharmaceutically acceptable salts and hydrates thereof.
