Abstract

An object is to enable LSM observation while correcting aberrations, as well as illumination with a desired pattern, using a single LCOS. An observation apparatus of the present invention includes an objective lens that focuses laser light emitted from a light source and radiates the laser light onto a specimen; an LCOS disposed at a position optically conjugate with the pupil position of the objective lens and capable of modulating, according to a wavefront modulation pattern, the wavefront of the laser light to be radiated onto the specimen by the objective lens; a scanner disposed at a position optically conjugate with the LCOS and capable of two-dimensionally scanning the laser light to be radiated onto the specimen by the objective lens; a focusing mechanism that adjusts the focal position, in the specimen, of the objective lens in the optical axis direction; and a control unit that controls the LCOS such that the LCOS corrects aberrations at the focal position adjusted by the focusing mechanism and forms a desired illumination pattern on the specimen.

Claims (18)

1. An observation apparatus comprising:

an objective lens that focuses illumination light emitted from a light source and radiates the illumination light onto a specimen;

a spatial-light-modulating element that is disposed at a position optically conjugate with a pupil position of the objective lens and is capable of modulating a wavefront of the illumination light to be radiated onto the specimen by the objective lens;

a scanning portion that is disposed at a position optically conjugate with the spatial-light-modulating element and is capable of two-dimensionally scanning the illumination light to be radiated onto the specimen by the objective lens;

a focusing portion that adjusts a focal position, in the specimen, of the objective lens in an optical axis direction; and

a control portion that controls the spatial-light-modulating element such that the spatial-light-modulating element corrects aberrations at the focal position and forms a predetermined illumination pattern on the specimen.

2. The observation apparatus according to claim 1 , wherein:

the objective lens, the scanning portion, and the spatial-light-modulating element are arranged on a same optical path, and

the control portion performs the control of the spatial-light-modulating element for forming the predetermined illumination pattern while the scanning of illumination light by the scanning portion is stopped.

The observation, apparatus according to claim 1 , further comprising a zero-order-light removing member disposed, in a manner capable of being inserted and removed, in an optical path between the spatial-light-modulating element and the scanning portion, at a position conjugate with the specimen, the zero-order-light removing member removing zero-order light contained in the illumination light having the wavefront thereof modulated by the spatial-light-modulating element, which is controlled by the control portion so as to form the predetermined illumination pattern.

4. The observation apparatus according to claim 1 , further comprising:

an optical-path-splitting portion that splits an optical path between the spatial-light-modulating element and the scanning portion into two; and

an optical-path-merging portion that merges, at a position between the scanning portion and the objective lens, the two optical paths split by the optical-path-splitting portion,

wherein the pupil position of the objective lens and the spatial-light-modulating element are located at positions optically conjugate with each other in each of the two optical paths.

The observation apparatus according to claim 4 , further comprising a zero-order-light removing member disposed at a position conjugate with the specimen, in the optical path split by the optical-path-splitting portion in which the scanning portion is not provided, the zero-order-light removing member removing zero-order light contained in the illumination light having the wavefront thereof modulated by the spatial-light-modulating element, which is controlled by the control portion so as to form the predetermined illumination pattern.

6. The observation apparatus according to claim 4 , wherein:

the optical-path-splitting portion is disposed at a position conjugate with the specimen and splits the optical path of the illumination light coming from the spatial-light-modulating element into an optical path of a portion of the illumination light containing the zero-order light and an optical path of a portion of the illumination light not containing the zero-order light, and

the scanning portion is disposed in the optical path of the portion of the illumination light containing the zero-order light.
