Abstract

Transposomes and oligonucleotide replacement methods to make DNA libraries that have distinct 5′ and 3′ tags, and to make directional libraries that are enriched for a desired strand.

Claims (98)

1. A method for making a transposome, comprising the steps of:

(a) providing a transposase; and

(b) providing a transferred end sequence comprising a nucleic acid comprising one or two copies of a transposase end sequence selected from the group consisting of:

(SEQ ID NO: 1)

MRWTGTGHWKAVGARACAV

and

(SEQ ID NO: 2)

NSHBGHSHDDRNGAKACAN,

but excluding

(SEQ ID NO: 3)

AGATGTGTATAAGAGACAG;

under conditions whereby the transposase binds to the transferred end sequence, thereby forming a transposome.

2. The method of claim 1 , wherein the nucleic acid is a transferred strand and comprises a tag sequence.

3. The method of claim 2 , wherein the tag sequence comprises one or more barcodes, one or more primer sites, one or more affinity tags, one or more reporter moieties, or any combination thereof.

4. The method of claim 1 , further comprising contacting the transposome with a target nucleic acid.

5. The method of claim 4 , wherein the contacting is performed in solution.

6. A transposome comprising a transposase and a nucleic acid comprising one or two copies of a transposase end sequence selected from the group consisting of:

but excluding

9. The transposome of claim 6 , wherein the nucleic acid is a transferred strand and comprises a tag sequence.

10. The transposome of claim 9 , wherein the tag sequence comprises one or more barcodes, one or more primer sites, one or more affinity tags, or one or more reporter moieties, or any combination thereof.

11. The transposome of claim 6 , further comprising a non-transferred strand that comprises:

12. The transposome of claim 6 , further comprising a non-transferred strand selected from the group consisting of: SEQ ID NO: 9, SEQ ID NO: 10, SEQ ID NO: 11, SEQ ID NO: 12, SEQ ID NO: 13, SEQ ID NO: 14, SEQ ID NO: 15, SEQ ID NO: 16, SEQ ID NO: 17, and SEQ ID NO: 18.

13. The transposome of claim 6 , wherein the transposome comprises a Tn5 or Mu transposase.

14. The transposome of claim 6 , wherein the transposome comprises a hyperactive Tn5 transposase.

15. A transposome comprising a transposase and a double-stranded nucleic acid comprising a transposase end sequence, the transposase end sequence comprising one or two copies of a first sequence comprising:

but excluding

16. The transposome of claim 15 , wherein the first sequence is selected from the group consisting of:

18. The transposome of claim 15 , wherein the second sequence is selected from the group consisting of: SEQ ID NO: 9, SEQ ID NO: 10, SEQ ID NO: 11, SEQ ID NO: 12, SEQ ID NO: 13, SEQ ID NO: 14, SEQ ID NO: 15, SEQ ID NO: 16, SEQ ID NO: 17, and SEQ ID NO: 18.

19. The transposome of claim 15 , wherein the transposome comprises a Tn5 or Mu transposase.

20. The transposome of claim 15 , wherein the transposome comprises a hyperactive Tn5 transposase.
