Methods for modulating the activity of insecticidal proteins
View Patent ↗Compositions and methods for controlling pests are disclosed. In particular, methods for modulating the activity of Cry1Ba proteins against lepidopteran or coleopteran pests are provided. Further, mutant Cry1Ba proteins having modulated insecticidal activity compared to native Cry1Ba proteins are disclosed.
1. A method of modulating the activity of a Cry1Ba protein, the method comprising introducing a mutation in a Cry1Ba protein at a position corresponding to or at amino acid position 150 of SEQ ID NO:2, wherein the resulting variant Cry1Ba protein has lower insecticidal activity against European corn borer ( Ostrinia nubilalis ) compared to a native Cry1Ba protein having a Tyr (Y) or His (H) at a position corresponding to or at amino acid position 150 of SEQ ID NO:2.
2. The method of claim 1 , wherein the mutation at the position corresponding to or at amino acid position 150 of SEQ ID NO:2 is Thr (T), Leu (L), Ala (A), Val (V), Ser (S), Arg (R), Gly (G), or Asp (D).
3. The method of claim 1 , wherein the mutation is introduced by mutating a Cry1Ba-encoding nucleic acid molecule at a codon encoding an amino acid corresponding to or at amino acid position 150 of SEQ ID NO:2.
4. The method of claim 3 , wherein the codon corresponds to or is the codon at positions 448-450 of SEQ ID NO:1.
5. The method of claim 4 , wherein the mutated codon encodes Thr (T), Leu (L), Ala (A), Val (V), Ser (S), Arg (R), Gly (G), or Asp (D).
6. A variant Cry1Ba protein resulting from the introduction of the mutation in claim 1 .
7. The variant Cry1Ba protein of claim 6 , wherein the mutation at the amino acid position corresponding to or at amino acid 150 of SEQ ID NO:2 is Thr (T), Leu (L), Ala (A), Val (V), Ser (S), Arg (R), Gly (G), or Asp (D).
