Abstract

Conjugates of a Factor VIII moiety and one or more water-soluble polymers are provided. Typically, the water-soluble polymer is poly(ethylene glycol) or a derivative thereof. Also provided are compositions comprising the conjugates, methods of making the conjugates, and methods of administering compositions comprising the conjugates to a patient.

Claims (22)

1. A monoPEGylated Factor VIII conjugate comprising a single poly(ethylene glycol) polymer covalently attached to a cysteine residue of a B-domain deleted Factor VIII polypeptide, wherein the single poly(ethylene glycol) polymer is branched and has a nominal average molecular weight in a range of about 20,000 daltons to about 85,000 daltons.

2. The monoPEGylated Factor VIII conjugate of claim 1 , wherein the single poly(ethylene glycol) polymer comprises PEG attached to a polyol core.

3. The monoPEGylated Factor VIII conjugate of claim 1 , wherein the single poly(ethylene glycol) polymer has a nominal average molecular weight in a range of about 53,000 daltons to about 85,000 daltons.

The monoPEGylated Factor VIII conjugate of claim 1 , wherein the single poly(ethylene glycol) polymer has a nominal average molecular weight selected from the group consisting of about 20,000 Daltons, about 22,500 Daltons, about 25,000 Daltons, about 30,000 Daltons, about 40,000 Daltons, about 50,000 Daltons, about 60,000 Daltons, and about 75,000 Daltons.

5. The monoPEGylated Factor VIII conjugate of claim 1 , wherein the cysteine residue to which the single poly(ethylene glycol) polymer is covalently attached is one that has been added to or substituted in the B-domain deleted Factor VIII polypeptide.

6. The monoPEGylated Factor VIII conjugate of claim 3 , wherein the cysteine residue to which the single poly(ethylene glycol) polymer is covalently attached is one that has been added to or substituted in the B-domain deleted Factor VIII polypeptide.

7. The monoPEGylated Factor VIII conjugate of claim 6 , wherein the added or substituted cysteine residue to which the single poly(ethylene glycol) polymer is covalently attached is located within the A1 subunit of the B-domain deleted Factor VIII polypeptide.

8. The monoPEGylated Factor VIII conjugate of claim 6 , wherein the added or substituted cysteine residue to which the single poly(ethylene glycol) polymer is covalently attached is located within the A2 subunit of the B-domain deleted Factor VIII polypeptide.

9. The monoPEGylated Factor VIII conjugate of claim 6 , wherein the added or substituted cysteine residue to which the single poly(ethylene glycol) polymer is covalently attached is located within the A3 subunit of the B-domain deleted Factor VIII polypeptide.

10. The monoPEGylated Factor VIII conjugate of claim 6 , wherein the added or substituted cysteine residue to which the single poly(ethylene glycol) polymer is covalently attached is located within the C1 subunit of the B-domain deleted Factor VIII polypeptide.

11. The monoPEGylated Factor VIII conjugate of claim 6 , wherein the added or substituted cysteine residue to which the single poly(ethylene glycol) polymer is covalently attached is located within the C2 subunit of the B-domain deleted Factor VIII polypeptide.

12. The monoPEGylated Factor VIII conjugate of claim 6 , wherein a thiol group of the cysteine residue is covalently attached to the single poly(ethylene glycol) polymer via reaction with an N-maleimidyl group of the poly(ethylene glycol) polymer.

The monoPEGylated Factor VIII conjugate of claim 12 , wherein the linkage between the single poly(ethylene glycol) polymer and the B-domain deleted Factor VIII polypeptide comprises, in addition to a thioether bond, a residue of an N-maleimidyl group to which the thioether sulfur is covalently attached.

14. The monoPEGylated Factor VIII conjugate of claim 13 , wherein the single poly(ethylene glycol) polymer comprises a series of atoms connecting the cysteine residue of the B-domain deleted Factor VIII polypeptide to a chain of repeating monomers within the single poly(ethylene glycol) polymer.

15. The monoPEGylated Factor VIII conjugate of claim 14 , wherein the series of atoms connecting the cysteine residue of the B-domain deleted Factor VIII polypeptide to the chain of repeating monomers within the single poly(ethylene glycol) polymer includes an amide group.

16. The monoPEGYlated Factor VIII conjugate of claim 14 , wherein the single poly(ethylene glycol) polymer comprises terminal methoxy capping groups.

17. The monoPEGylated Factor VIII conjugate of claim 16 , wherein the conjugate is bioactive.

18. The monoPEGylated Factor VIII conjugate of claim 1 , wherein the single poly(ethylene glycol) polymer is covalently attached to the cysteine residue of the B-domain deleted Factor VIII polypeptide via a thioether bond.

19. The monoPEGylated Factor VIII conjugate of claim 1 , wherein the single poly(ethylene glycol) polymer has a nominal average molecular weight of about 60,000 daltons.

20. The monoPEGylated Factor VIII conjugate of claim 3 , wherein the cysteine residue to which the single poly(ethylene glycol) polymer is covalently attached is located within the A3 subunit of the B-domain deleted Factor VIII polypeptide.

21. The monoPEGylated Factor VIII conjugate of claim 20 , wherein the single poly(ethylene glycol) polymer has a nominal average molecular weight of about 60,000 daltons.

22. The monoPEGylated Factor VIII conjugate of claim 21 , wherein the cysteine residue to which the single poly(ethylene glycol) polymer is covalently attached is one that has been added to or substituted in the B-domain deleted Factor VIII polypeptide.
