Abstract

Improved oral dosage forms of methyl hydrogen fumarate and prodrugs thereof are disclosed. Methods of treating diseases such as multiple sclerosis and psoriasis using such dosage forms are also disclose.

Claims (21)

1. An oral dosage form comprising:

a core comprising a therapeutically effective amount of (N,N-Diethylcarbamoyl)methyl methyl(2E)but-2-ene-1,4-dioate;

a barrier layer directly coating said core, the barrier layer comprising a non-ionizable polymer; the barrier layer preventing direct contact between the core and an enteric coating; and

the enteric coating directly surrounding said barrier layer and comprising an enteric polymer having carboxylic acid moieties, the enteric polymer (i) being soluble in aqueous solutions having a pH above 7.5 and (ii) starting to become soluble in an aqueous solution at a pH in the range of 4.5 to 7.5.

2. The oral dosage form of claim 1 , wherein the barrier layer material is selected from non-ionizable cellulosic polymers, non-ionizable vinyl polymers, and non-ionizable polyvinyl alcohol polymers.

3. The oral dosage form of claim 1 , wherein the barrier layer material is a sugar.

4. The oral dosage form of claim 1 , wherein the barrier layer comprises at least 5 wt % of the coated core for cores having a size of 2 mm or less.

5. The oral dosage form of claim 1 , wherein the barrier layer comprises at least 0.5 wt % of the coated core for cores having a size greater than 6 mm.

6. The oral dosage form of claim 1 , wherein the barrier layer has an average thickness of at least 5 μm.

7. The oral dosage form of claim 1 , wherein the barrier layer has an average thickness of at least 15 μm.

8. The oral dosage form of claim 1 , wherein the core is a compressed tablet and the barrier layer and enteric coating are each a coating on the tablet.

9. The oral dosage form of claim 8 , wherein the tablet contains from 100 to 1200 mg of (N,N-Diethylcarbamoyl)methyl methyl(2E)but-2-ene-1,4-dioate and one or more tableting excipients.

10. The oral dosage form of claim 9 , wherein the tableting excipients are selected from binders, diluents, disintegrants, glidiants and lubricants.

11. The oral dosage form of claim 1 , wherein the dosage form is a capsule containing a multiplicity of said enteric coated cores.

12. The oral dosage form of claim 1 , wherein the core comprises an immediate release dosage form.

13. The oral dosage form of claim 1 , wherein the core comprises a sustained release dosage form.

14. The oral dosage form of claim 1 , wherein the enteric polymer is selected from methyacrylic acid polymers, cellulose acetate phthalate polymers, hydroxypropylmethyl cellulose acetate succinate polymers, hydroxypropylmethyl cellulose phthalate polymers and polyvinyl acetate phthalate polymers.

15. A method of treating a disease selected from the group consisting of multiple sclerosis and psoriasis in a patient, comprising orally administering to a patient in need thereof a dosage form of claim 1 .

16. The method of claim 15 , wherein the oral administration is sufficient to obtain a therapeutic concentration of methyl hydrogen fumarate in blood plasma of the patient of at least 0.7 μg/ml at a time within 24 hours after said oral administration.

17. The method of any one of claim 15 , wherein the oral administration is sufficient to obtain an area under a concentration of methyl hydrogen fumarate in the blood plasma versus time curve (AUC) of at least 12.0 μg·hr/ml over 24 hours after start of the oral administration.

The oral dosage form of claim 14 , wherein the enteric coating is selected form the group consisting of hydroxypropyl methyl cellulose acetate succinate, hydroxypropyl methyl cellulose succinate, hydroxypropyl cellulose acetate succinate, hydroxyethyl methyl cellulose succinate, hydroxyethyl cellulose acetate succinate, hydroxypropyl methyl cellulose phthalate, hydroxyethyl methyl cellulose acetate succinate, hydroxyethyl methyl cellulose acetate phthalate, carboxymethyl cellulose, carboxymethyl cellulose, cellulose acetate phthalate, methyl cellulose acetate phthalate, ethyl cellulose acetate phthalate, hydroxypropyl cellulose acetate phthalate, hydroxypropyl methyl cellulose acetate succinate phthalate, hydroxypropyl cellulose succinate phthalate succinate, hydroxypropyl methyl cellulose acetate succinate phthalate, hydroxypropyl methyl cellulose succinate phthalate, cellulose propionate phthalate, hydroxypropyl cellulose butyrate phthalate, cellulose acetate trimellitate, methyl cellulose acetate trimellitate, ethyl cellulose acetate trimellitate, hydroxypropyl cellulose acetate trimellitate, hydroxypropyl methyl cellulose acetate trimellitate, hydroxypropyl cellulose acetate trimellitate succinate, cellulose propionate trimellitate, cellulose butyrate trimellitate, cellulose acetate terephthalate, cellulose acetate isophthalate, cellulose acetate pyridinedicarboxylate, salicylic acid cellulose acetate, hydroxypropyl salicylic acid cellulose acetate, ethylbenzoic acid cellulose acetate, hydroxypropyl ethylbenzoic acid cellulose acetate, ethyl phthalic acid cellulose acetate, ethyl nicotinic acid cellulose acetate, and ethyl picolinic acid cellulose acetate.
