RNA interference in skin indications
The present invention relates to RNAi constructs with improved tissue and cellular uptake characteristics and methods of use of these compounds in dermal applications.
1. A method for treating or preventing a fibrotic disorder in a subject in need thereof, the method comprising:
administering to the subject a therapeutically effective amount for treating or preventing a fibrotic disorder of a double stranded nucleic acid molecule comprising a guide strand, with a minimal length of 16 nucleotides, and a passenger strand forming a double stranded nucleic acid, having a double stranded region and a single stranded region, the double stranded region having 8-15 nucleotides in length, the single stranded region having 4-12 nucleotides in length, wherein the single stranded region comprises 2-12 phosphorothioate modifications, wherein at least 40% of the nucleotides of the double stranded nucleic acid molecule are modified, and wherein the double stranded nucleic acid molecule is directed against a gene encoding-TGFB1 or TGFB2.
2. The method of claim 1 , wherein the double stranded nucleic acid molecule is administered via intradermal injection.
3. The method of claim 1 , wherein the double stranded nucleic acid molecule is administered via local administration to the skin.
4. The method of claim 1 , wherein the double stranded nucleic acid molecule is administered on the skin of the subject in need thereof.
5. The method of claim 4 , wherein the double stranded nucleic acid molecule is in the form of a cream or ointment.
6. The method of claim 1 , wherein the double stranded nucleic acid molecule is administered by local injection.
7. The method of claim 1 , wherein the fibrotic disorder is selected from the group consisting of pulmonary fibrosis, liver cirrhosis, scleroderma, glomerulonephritis, lung fibrosis, liver fibrosis, skin fibrosis, muscle fibrosis, radiation fibrosis, kidney fibrosis, proliferative vitreoretinopathy, and uterine fibrosis.
8. The method of claim 1 , wherein the single stranded region contains 3, 4, 5, 6, 7, 8, 9, 10, 11 or 12 phosphorothioate modifications.
9. The method of claim 1 , wherein the double stranded nucleic acid molecule comprises a hydrophobic conjugate attached to the guide strand or passenger strand.
10. The method of claim 9 , wherein the conjugate is cholesterol.
11. The method of claim 1 , wherein at least one of the nucleotides of the isolated double stranded nucleic acid molecule that is modified comprises a 2′ O-methyl or a 2′-fluoro modification.
12. The method of claim 1 , wherein at least one of the nucleotides of the isolated double stranded nucleic acid molecule that is modified comprises a hydrophobic modification.
13. The method of claim 1 , wherein the double stranded region is 11, 12, 13, or 14 nucleotides long.
14. The method of claim 1 , wherein each nucleotide within the single stranded region has a phosphorothioate modification.
15. The method of claim 9 , wherein the hydrophobic conjugate is attached to the double stranded nucleic acid molecule through a linker.
16. The method of claim 15 , wherein the linker is a TEG linker.
17. The method of claim 1 , wherein the single stranded region is at least 4, at least 5, or at least 6 nucleotides long.
18. A method for treating or preventing a fibrotic disorder in a subject in need thereof, the method comprising:
administering to the subject a therapeutically effective amount for treating or preventing a fibrotic disorder of a double stranded nucleic acid molecule comprising a guide strand, with a minimal length of 16 nucleotides, and a passenger strand forming a double stranded nucleic acid, having a double stranded region and a single stranded region, the double stranded region having 8-15 nucleotides in length, the single stranded region having 4-12 nucleotides in length, wherein the single stranded region comprises 2-12 phosphorothioate modifications, wherein at least 40% of the nucleotides of the double stranded nucleic acid molecule are modified, and wherein the double stranded nucleic acid molecule is directed against a gene encoding SPP1.
19. A method for treating or preventing a fibrotic disorder in a subject in need thereof, the method comprising:
administering to the subject a therapeutically effective amount for treating or preventing a fibrotic disorder of a double stranded nucleic acid molecule comprising a guide strand, with a minimal length of 16 nucleotides, and a passenger strand forming a double stranded nucleic acid, having a double stranded region and a single stranded region, the double stranded region having 8-15 nucleotides in length, the single stranded region having 4-12 nucleotides in length, wherein the single stranded region comprises 2-12 phosphorothioate modifications, wherein at least 40% of the nucleotides of the double stranded nucleic acid molecule are modified, and wherein the double stranded nucleic acid molecule is directed against a gene encoding PTGS2.
