Abstract

Systems and methods are provided for loading a drug delivery device. In one embodiment, the method includes positioning one or more solid drug units upstream of the drug delivery device; and driving the drug units into the drug delivery device with a flow of pressurized gas. The systems and methods may be used to load a drug delivery device that includes a device body, a number of solid, compressed drug tablets, and a retention frame. The device body includes a drug reservoir lumen and a retention frame lumen. The number of solid, compressed drug tablets is positioned in the drug reservoir lumen, and the retention frame is positioned in the retention frame lumen.

Claims (22)

1. A method for administration of a drug into the urinary bladder of a patient, the method comprising:

deploying into the urinary bladder of the patient a drug delivery device which comprises (i) elastic walls defining a drug reservoir lumen and a retention frame lumen, (ii) a plurality of solid drug tablets positioned in the drug reservoir lumen, and (iii) a retention frame positioned in the retention frame lumen, the retention frame comprising a shape memory material set in a coiled retention shape, wherein the device is elastically deformable between a relatively lower-profile shape to permit insertion of the device through the patient's urethra and a relatively higher-profile coiled shape for retention of the device within the patient's urinary bladder; and then

releasing solubilized drug from the device into the patient's urinary bladder,

wherein the device comprise a drug reservoir tube that defines the drug reservoir lumen; and a retention frame tube that defines the retention frame lumen, the retention frame tube being longitudinally aligned with and coupled to the drug reservoir tube along their entire lengths, wherein the drug reservoir tube and the retention frame tube are each substantially cylindrical and together form a monolithic structure.

2. The method of claim 1 , wherein urine enters the drug reservoir lumen in vivo and dissolves the solid drug tablets to produce the solubilized drug.

3. The method of claim 1 , wherein the releasing of the solubilized drug comprises dispensing the solubilized drug out of the drug reservoir lumen through one or more apertures in the elastic walls, driven by osmotic pressure in the drug reservoir lumen.

4. The method of claim 1 , wherein the elastic walls defining the drug reservoir lumen have no apertures and releasing of the solubilized drug comprises diffusion of the solubilized drug through at least one of the elastic walls.

The method of claim 1 , wherein the deploying comprises passing the drug delivery device through a working channel of catheter, cystoscope, or other deployment instrument positioned in patient's urethra, and wherein the drug delivery device transitions to the higher-profile coiled shape upon emerging from the deployment instrument into the patient's urinary bladder.

6. The method of claim 1 , wherein the drug tablets are mini-tablets aligned in the drug reservoir lumen.

7. The method of claim 1 , wherein the drug reservoir and retention frame tubes comprise a water permeable silicone.

8. The method of claim 1 , wherein the retention frame comprises an elastic wire.

9. The method of claim 8 , wherein the elastic wire comprises a nickel-titanium alloy or another superelastic alloy.

10. The method of claim 1 , wherein the elastic walls defining the drug reservoir lumen and the retention frame lumen are non-resorbable.

The method of claim 1 , wherein interstices between the drug tablets permit deformation of the device body when the device is elastically deformed from the relatively lower-profile shape during insertion of the device through the patient's urethra to the relatively higher-profile coiled shape within the patient's urinary bladder.

12. The method of claim 1 , wherein the plurality of solid drug tablets comprises a single drug.

13. The method of claim 1 , wherein the solid drug tablets having a length:diameter aspect ratio greater than 1:1.

14. A method for administration of a drug into the urinary bladder of a patient, the method comprising:

deploying into the urinary bladder of the patient a drug delivery device which comprises (i) elastic walls defining a drug reservoir lumen and a retention frame lumen, (ii) a plurality of solid drug tablets positioned in the drug reservoir lumen, and (iii) a retention frame positioned in the retention frame lumen, the retention frame comprising a shape memory material set in a coiled retention shape, wherein the device is elastically deformable between a relatively lower-profile shape to permit insertion of the device through the patient's urethra and a relatively higher-profile coiled shape for retention of the device within the patient's urinary bladder; and then

releasing solubilized drug from the device into the patient's urinary bladder,

wherein the solid drug tablets are cylindrical in shape and have opposed flat ends.

15. The method of claim 14 , wherein the plurality of solid drug tablets are positioned end-to-end in the drug reservoir lumen and the drug tablets having a length:width aspect ratio greater than 1:1.

16. The method of claim 15 , wherein each of the solid drug tablets has a diameter from 1.0 to 3.2 mm and a length from 1.7 to 4.8 mm.
