Modulators and methods of use
View Patent ↗Novel modulators, including antibodies and derivatives thereof, and methods of such modulators to treat hyperproliferative disorders are provided.
1. A method of treating an EFNA4-expressing neoplastic disorder comprising administering a therapeutically effective amount of a composition comprising an antibody or antigen-binding fragment thereof that specifically binds to EFNA4 to a subject in need thereof, wherein the antibody or antigen-binding fragment thereof comprises at least one heavy chain variable region and at least one light chain variable region, wherein the at least one heavy chain variable region comprises three CDRs of a heavy chain variable region set forth as SEQ ID NO: 157 and wherein the at least one light chain variable region comprises three CDRs of a light chain variable region set forth as SEQ ID NO: 159.
2. The method of claim 1 , wherein the neoplastic disorder comprises a solid tumor.
3. The method of claim 1 , wherein the neoplastic disorder is breast cancer, ovarian cancer, colorectal cancer, liver cancer, or lung cancer.
4. The method of claim 3 , wherein the neoplastic disorder is breast cancer.
5. The method of claim 3 , wherein the neoplastic disorder is ovarian cancer.
6. The method of claim 1 , wherein the neoplastic disorder is a hematologic malignancy.
7. The method of claim 6 , wherein the hematologic malignancy is leukemia.
8. The method of claim 1 , wherein the antibody or antigen-binding fragment thereof comprises at least one heavy chain variable region and at least one light chain variable region, wherein the at least one heavy chain variable region comprises three CDRs set forth as residues 31-35 of SEQ ID NO: 157 for CDR-H1, residues 50-65 of SEQ ID NO: 157 for CDR-H2, and residues 95-102 of SEQ ID NO: 157 for CDR-H3, and wherein the at least one light chain variable region comprises three CDRs set forth as residues 24-34 of SEQ ID NO: 159 for CDR-L1, residues 50-56 of SEQ ID NO: 159 for CDR-L2, and residues 89-97 of SEQ ID NO: 159 for CDR-L3, wherein the residues are numbered according to Kabat.
9. The method of claim 1 , wherein the antibody or antigen-binding fragment thereof comprises at least one heavy chain variable region and at least one light chain variable region, wherein the at least one heavy chain variable region comprises three CDRs set forth as residues 26-32 of SEQ ID NO: 157 for CDR-H1, residues 53-55 of SEQ ID NO: 157 for CDR-H2, and residues 96-101 of SEQ ID NO: 157 for CDR-H3, and wherein the at least one light chain variable region comprises three CDRs set forth as residues 26-32 of SEQ ID NO: 159 for CDR-L1, residues 50-52 of SEQ ID NO: 159 for CDR-L2, and residues 91-96 of SEQ ID NO: 159 for CDR-L3, wherein the residues are numbered according to Chothia.
10. The method of claim 1 , wherein the antibody or antigen-binding fragment thereof comprises at least one heavy chain variable region and at least one light chain variable region, wherein the at least one heavy chain variable region comprises three CDRs set forth as residues 30-35 of SEQ ID NO: 157 for CDR-H1, residues 47-58 of SEQ ID NO: 157 for CDR-H2, and residues 93-101 of SEQ ID NO: 157 for CDR-H3, and wherein the at least one light chain variable region comprises three CDRs set forth as residues 30-36 of SEQ ID NO: 159 for CDR-L1, residues 46-55 of SEQ ID NO: 159 for CDR-L2, and residues 89-96 of SEQ ID NO: 159 for CDR-L3, wherein the residues are numbered according to MacCallum.
11. The method of claim 1 , wherein the antibody or antigen-binding fragment thereof comprises a heavy chain variable region set forth as SEQ ID NO: 157 and a light chain variable region set forth as SEQ ID NO: 159.
12. The method of claim 1 , wherein the antibody or antigen-binding fragment thereof is conjugated, linked or otherwise associated with a cytotoxic agent.
13. The method of claim 11 , wherein the antibody or antigen-binding fragment thereof is conjugated, linked or otherwise associated with a cytotoxic agent.
14. A method of treating an EFNA4-expressing cancer comprising administering a therapeutically effective amount of a composition comprising an antibody or antigen-binding fragment thereof that specifically binds to EFNA4 to a subject in need thereof, wherein the antibody or antigen-binding fragment thereof comprises at least one heavy chain variable region and at least one light chain variable region, wherein the at least one heavy chain variable region comprises three CDRs of a heavy chain variable region set forth as SEQ ID NO: 157 and wherein the at least one light chain variable region comprises three CDRs of a light chain variable region set forth as SEQ ID NO: 159.
15. The method of claim 14 , wherein the cancer comprises a solid tumor.
16. The method of claim 14 , wherein the cancer is breast cancer, ovarian cancer, colorectal cancer, liver cancer, or lung cancer.
17. The method of claim 16 , wherein the cancer is breast cancer.
18. The method of claim 16 , wherein the cancer is ovarian cancer.
19. The method of claim 14 , wherein the cancer is a hematologic malignancy.
20. The method of claim 19 , wherein the hematologic malignancy is leukemia.
21. The method of claim 14 , wherein the antibody or antigen-binding fragment thereof comprises at least one heavy chain variable region and at least one light chain variable region, wherein the at least one heavy chain variable region comprises three CDRs set forth as residues 31-35 of SEQ ID NO: 157 for CDR-H1, residues 50-65 of SEQ ID NO: 157 for CDR-H2, and residues 95-102 of SEQ ID NO: 157 for CDR-H3, and wherein the at least one light chain variable region comprises three CDRs set forth as residues 24-34 of SEQ ID NO: 159 for CDR-L1, residues 50-56 of SEQ ID NO: 159 for CDR-L2, and residues 89-97 of SEQ ID NO: 159 for CDR-L3, wherein the residues are numbered according to Kabat.

The method of claim 14 , wherein the antibody or antigen-binding fragment thereof comprises at least one heavy chain variable region and at least one light chain variable region, wherein the at least one heavy chain variable region comprises three CDRs set forth as residues 26-32 of SEQ ID NO: 157 for CDR-H1, residues 53-55 of SEQ ID NO: 157 for CDR-H2, and residues 96-101 of SEQ ID NO: 157 for CDR-H3, and wherein the at least one light chain variable region comprises three CDRs set forth as residues 26-32 of SEQ ID NO: 159 for CDR-L1, residues 50-52 of SEQ ID NO: 159 for CDR-L2, and residues 91-96 of SEQ ID NO: 159 for CDR-L3, wherein the residues are numbered according to Chothia.

The method of claim 14 , wherein the antibody or antigen-binding fragment thereof comprises at least one heavy chain variable region and at least one light chain variable region, wherein the at least one heavy chain variable region comprises three CDRs set forth as residues 30-35 of SEQ ID NO: 157 for CDR-H1, residues 47-58 of SEQ ID NO: 157 for CDR-H2, and residues 93-101 of SEQ ID NO: 157 for CDR-H3, and wherein the at least one light chain variable region comprises three CDRs set forth as residues 30-36 of SEQ ID NO: 159 for CDR-L1, residues 46-55 of SEQ ID NO: 159 for CDR-L2, and residues 89-96 of SEQ ID NO: 159 for CDR-L3, wherein the residues are numbered according to MacCallum.
24. The method of claim 14 , wherein the antibody or antigen-binding fragment thereof comprises a heavy chain variable region set forth as SEQ ID NO: 157 and a light chain variable region set forth as SEQ ID NO: 159.
25. The method of claim 14 , wherein the antibody or antigen-binding fragment thereof is conjugated, linked or otherwise associated with a cytotoxic agent.
26. The method of claim 24 , wherein the antibody or antigen-binding fragment thereof is conjugated, linked or otherwise associated with a cytotoxic agent.
27. A method of treating an EFNA4-expressing neoplastic disorder comprising administering a therapeutically effective amount of a composition comprising an antibody or antigen-binding fragment thereof that specifically binds to EFNA4 to a subject in need thereof, wherein the antibody or antigen-binding fragment thereof comprises three CDRs of a heavy chain variable region set forth as SEQ ID NO: 157 and three CDRs of a light chain variable region set forth as SEQ ID NO: 159, wherein said EFNA4-expressing neoplastic disorder is breast cancer, ovarian cancer, colorectal cancer, liver cancer, or lung cancer.
