Abstract

The invention provides methods for assessing one or more predetermined characteristics or properties of a microfluidic droplet within a microfluidic channel, and regulating one or more fluid flow rates within that channel to selectively alter the predetermined microdroplet characteristic or property using a feedback control.

Claims (20)

1. A method of merging droplets, comprising

flowing a first plurality of fluidic droplets within a microfluidic channel at a first frequency;

providing a second plurality of fluidic droplets within the microfluidic channel at a second frequency;

capturing an image of the first plurality and/or second plurality of fluidic droplets over a plurality of pulses of illumination;

assessing the first frequency and/or the second frequency from the captured image;

transmitting the assessment of the first and/or the second frequencies to a feedback controller, wherein the feedback controller adjusts a flow rate of one or more fluids to provide a desired frequency ratio of the first plurality of fluidic droplets to said second plurality of fluidic droplets; and

merging at least one of the first plurality of fluidic droplets with at least one of the second plurality of fluidic droplets within the microfluidic channel.

2. The method of claim 1 , wherein the first plurality of fluidic droplets each comprise a biological library element.

3. The method of claim 1 , wherein the second plurality of fluidic droplets each comprise a single primer pair.

4. The method of claim 1 , wherein the second plurality of fluidic droplets are pre-made library of droplets.

5. The method of claim 1 , wherein the second plurality of fluidic droplets each comprise at least a different element.

6. The method of claim 1 , wherein the second plurality of fluidic droplets each comprise a different antibody species.

7. The method of claim 1 , wherein the first plurality of fluidic droplets each comprise a cell.

8. The method of claim 1 , wherein the first plurality of fluidic droplets each comprise an enzyme.

9. The method of claim 1 , wherein the first plurality of droplets are flowing in a carrier fluid.

10. The method of claim 9 , wherein the drive fluid comprise the same substance as the carrier fluid.

11. The method of claim 1 , wherein the second plurality of fluidic droplet is provided at a frequency of 900 to 1100 per second.

12. The method of claim 9 , wherein the feedback controller adjusts the flow rate of the carrier fluid.

13. The method of claim 10 , wherein the feedback controller adjusts the flow rate of the drive fluid.

14. The method of claim 2 , wherein the biological library element is selected from the group consisting of compounds, primer pairs, and antibody species.
