Abstract

In alternative embodiments, the invention provides compositions, e.g., formulations, used for gastric, gastrointestinal and/or colonic treatments or lavage, e.g., orthostatic lavage, e.g., for inducing the purgation (e.g., cleansing) of a gastrointestinal (GI) tract, including a colon; and methods for making and using them. In alternative embodiments, compositions and methods of the invention are used for the stabilization, amelioration, treatment and/or prevention of constipation, for the treatment of abdominal pain, particularly non-specific abdominal pain, and diarrhea, including diarrhea caused by a drug side effect, a psychological condition, a disease or a condition such as Crohn's Disease, a poison, a toxin or an infection, e.g., a toxin-mediated traveler's diarrhea, or C. difficile or the pseudo-membranous colitis associated with this infection. In alternative embodiments, the invention provides pharmaceuticals and products (articles) of manufacture for delivering these compositions and formulations to an individual, e.g., a human or an animal. The invention also provides devices for delivering a fecal material to a patient.

Claims (24)

1. A pharmaceutical composition comprising an added cryoprotectant and extracted stool bacterial material without fiber.

2. The pharmaceutical composition of claim 1 , wherein said composition further comprises an antioxidant.

3. The pharmaceutical composition of claim 1 , wherein said stool composition further comprises cysteine.

4. A filtered and homogenized stool composition comprising the substantially entire microbiota of a stool and an added antioxidant, wherein said stool composition is free of rough particulate matter of said stool.

5. The filtered and homogenized stool composition of claim 4 , wherein said antioxidant comprises cysteine.

6. The filtered and homogenized stool composition of claim 4 , wherein said stool composition further comprises a cryoprotectant.

7. The filtered and homogenized stool composition of claim 6 , wherein said cryoprotectant comprises trehalose.

8. The filtered and homogenized stool composition of claim 7 , wherein said antioxidant comprises cysteine.

9. The filtered and homogenized stool composition of claim 8 , wherein said stool composition is in a lyophilized form.

10. The filtered and homogenized stool composition of claim 4 , wherein said stool composition is in a lyophilized form.

11. A capsule adapted for intestinal delivery comprising a filtered and homogenized stool composition comprising the substantially entire microbiota of a stool and free of rough particulate matter of said stool.

12. The capsule adapted for intestinal delivery of claim 11 , wherein said stool composition comprises an oxygen scavenging material.

13. The capsule adapted for intestinal delivery of claim 11 , wherein said stool composition is free of non-living fecal matter.

14. The capsule adapted for intestinal delivery of claim 11 , wherein said stool composition further comprises a cryoprotectant.

15. The capsule adapted for intestinal delivery of claim 11 , wherein said stool composition further comprises glycerol, skim milk, erythritol, arabitol, sorbitol, glucose, or fructose.

16. The capsule adapted for intestinal delivery of claim 11 , wherein said stool composition further comprises polyethylene glycol.

17. The capsule adapted for intestinal delivery of claim 11 , wherein said stool composition further comprises a Bacteroides species, a Firmicutes species, Bacillus thuringiensis , or any combination thereof, from a culture.

18. The capsule adapted for intestinal delivery of claim 11 , wherein said stool composition is in a liquid, frozen, spray-dried, lyophilized, or powder form.

19. The capsule adapted for intestinal delivery of claim 11 , wherein said stool composition is in a liquid form.

20. The capsule adapted for intestinal delivery of claim 11 , wherein said stool composition is in a lyophilized form.

21. The capsule adapted for intestinal delivery of claim 11 , wherein said stool composition is formulated with or is mixed with a saline, a defoaming agent, a surfactant agent, a lubricant, an acid neutralizer, a marker, a cell marker, a drug, an antibiotic, a contrast agent, a dispersal agent, a buffer or a buffering agent, a sweetening agent, a debittering agent, a flavoring agent, a pH stabilizer, an acidifying agent, a preservative, a desweetening agent, coloring agent, at least one vitamin, mineral supplement, a dietary supplement, a prebiotic nutrient or any combination thereof.

22. The capsule adapted for intestinal delivery of claim 11 , wherein said stool composition is capable of restoring a patient's colonic flora and ameliorating, stabilizing, or treating a dysfunctional bowel.

23. The capsule adapted for intestinal delivery of claim 11 , wherein said stool composition is encapsulated as an enteric-coated capsule.

24. The capsule adapted for intestinal delivery of claim 23 , wherein said enteric-coated capsule is a graded release capsule.
