Abstract

A breath-powered, dry powder inhaler, a cartridge, and a pulmonary drug delivery system are provided. The dry powder inhaler can be provided with or without a unit dose cartridge for using with the inhaler. The inhaler and/or cartridge can be provided with a drug delivery formulation comprising, for example, a diketopiperazine and an active ingredient, including, peptides and proteins such as insulin and glucagon-like peptide 1 for the treatment of diabetes and/or obesity. The dry powder inhaler is compact; can be provided in various shapes and sizes, colors, and comprises a housing, a mouthpiece, a cartridge placement area, and a mechanism for opening and closing the medicament cartridge. The device is easy to manufacture, provides a pre-metered single unit dose, it is relatively easy to use, and can be reusable or disposable.

Claims (26)

1. A medicament cartridge for an inhaler, comprising:

a cartridge disk including two or more containers configured to hold a medicament;

a lid including one or more inlets and one or more dispensing ports for each container; and

a seal disk including one aperture that is configured to align with a selected one of the one or more dispensing ports and an opening configured to align with one of the one or more inlets,

wherein the one aperture is configured to transition from one container to another container when the cartridge disk is rotated, wherein the lid includes a top for each container that moves by a translational motion relative to the container.

2. The medicament cartridge of claim 1 , wherein the medicament comprises an active ingredient.

The medicament cartridge of claim 2 , wherein the active ingredient is a hormone, an anticoagulant, an immunomodulating agent, a vaccine, a cytotoxic agent, an antibiotic, a vasoactive agent, a neuroactive agent, an anaesthetic, sedative, a steroid, a decongestant, an antiviral, an antisense, an antigen, or an antibody.

The medicament cartridge of claim 2 , wherein the active ingredient is insulin, heparin, calcitonin, felbamate, sumatriptan, parathyroid hormone, growth hormone, erythropoietin, AZT, DDI, granulocyte macrophage colony stimulating factor, lamotrigine, chorionic gonadotropin releasing factor, luteinizing releasing hormone, beta-galactosidase, exendin, vasoactive intestinal peptide, follicle stimulating hormone, vasoactive intestinal peptide, parathyroid hormone, parathyroid hormone related protein, glucagon-like peptide-1, exendin, oxyntomodulin, peptide YY, interleukin 2-inducible tyrosine kinase, Bruton's tyrosine kinase, inositol-requiring kinase 1, PC-DAC-modified derivative, or O-glycosylated forms of PC-DAC, parathyroid hormone 1-34, argatroban, or a combination thereof.

5. The medicament cartridge of claim 4 , wherein the active ingredient is insulin.

6. The medicament cartridge of claim 4 , wherein the active ingredient is oxyntomodulin.

7. The medicament cartridge of claim 4 , wherein the active ingredient is glucagon-like peptide-1.

8. The medicament cartridge of claim 1 , wherein the medicament comprises a di ketopiperazine.

9. The medicament cartridge of claim 8 , wherein the diketopiperazine is

10. A dry powder medicament cartridge for an inhaler, comprising:

a cartridge disk including two or more containers configured to hold a medicament;

a lid including one or more inlets and one or more dispensing ports for each container, wherein the one or more dispensing ports are configured to have a smallest dimension less than 3 mm to exclude release of larger dry powder aggregates; and

a seal disk including one aperture that is configured to align with a selected one of the one or more dispensing ports and an opening configured to align with one of the one or more inlets,

wherein the one aperture is configured to transition from one container to another container when the cartridge disk is rotated, wherein the lid includes a top for each container that moves by a translational motion relative to the container.

11. The dry powder medicament cartridge of claim 10 , wherein the medicament comprises an active ingredient.

The dry powder medicament cartridge of claim 11 , wherein the active ingredient is hormone, an anticoagulant, an immunomodulating agent, a vaccine, a cytotoxic agent, an antibiotic, a vasoactive agent, a neuroactive agent, an anaesthetic, sedative, a steroid, a decongestant, an antiviral, an antisense, an antigen, or an antibody.

13. The dry powder medicament cartridge of claim 11 , wherein the active ingredient is insulin, heparin, calcitonin, felbamate, sumatriptan, parathyroid hormone, growth hormone, erythropoietin, AZT, DDI, granulocyte macrophage colony stimulating factor, lamotrigine, chorionic gonadotropin releasing factor, luteinizing releasing hormone, beta-galactosidase, exendin, vasoactive intestinal peptide, follicle stimulating hormone, vasoactive intestinal peptide, parathyroid hormone, parathyroid hormone related protein, glucagon-like peptide-1, exendin, oxyntomodulin, peptide YY, interleukin 2-inducible tyrosine kinase, Bruton's tyrosine kinase, inositol-requiring kinase 1, PC-DAC-modified derivative, or O-glycosylated forms of PC-DAC, parathyroid hormone 1-34, argatroban, or a combination thereof.

14. The dry powder medicament cartridge of claim 13 , wherein the active ingredient is insulin.

15. The dry powder medicament cartridge of claim 13 , wherein the active ingredient is oxyntomodulin.

16. The dry powder medicament cartridge of claim 13 , wherein the active ingredient is glucagon-like peptide-1.

17. The dry powder medicament cartridge of claim 10 , wherein the medicament comprises a diketopiperazine.
