Caninized antibodies
The present invention discloses caninized antibodies with specific properties. The present invention also discloses caninized murine antibodies againstcanine PD-1 that have a high binding affinity for canine PD-1. The invention further discloses the use of the caninized antibodies of the present invention in the treatment of cancer in dogs.
1. A canine fragment crystallizable region (cFc region) that comprises the amino acid sequence of SEQ ID NO: 130
that comprises the substituted amino acid residues of P4A, D31A, N63A, A93G, and P95A.
2. A caninized antibody that comprises the cFc region of claim 1 .
3. The caninized antibody of claim 2 , that further comprises a heavy chain complementary determining region 1 (VH CDR1) that comprises the amino acid sequence of SEQ ID NO: 29; a heavy chain complementary determining region 2 (VH CDR2) that comprises the amino acid sequence of SEQ ID NO: 33; a heavy chain complementary determining region 3 (VH CDR3) that comprises the amino acid sequence of SEQ ID NO: 38; a light chain complementary determining region 1 (VL CDR1) that comprises the amino acid sequence of SEQ ID NO: 15; a light chain complementary determining region 2 (VL CDR2) that comprises the amino acid sequence of SEQ ID NO: 18; and a light chain complementary determining region 3 (VL CDR3) that comprises the amino acid sequence of SEQ ID NO: 24.
4. The caninized antibody of claim 2 , that further comprises a heavy chain complementary determining region 1 (VH CDR1) that comprises the amino acid sequence of SEQ ID NO: 27; a heavy chain complementary determining region 2 (VH CDR2) that comprises the amino acid sequence of SEQ ID NO: 31; a heavy chain complementary determining region 3 (VH CDR3) that comprises the amino acid sequence of SEQ ID NO: 36; a light chain complementary determining region 1 (VL CDR1) that comprises the amino acid sequence of SEQ ID NO: 13; a light chain complementary determining region 2 (VL CDR2) that comprises the amino acid sequence of SEQ ID NO: 19; and a light chain complementary determining region 3 (VL CDR3) that comprises the amino acid sequence of SEQ ID NO: 25.
5. A canine fragment crystallizable region (cFc region) that comprises the amino acid sequence of SEQ ID NO: 130 that comprises the substituted amino acids residues of D31A and N63A.
6. A caninized antibody that comprises the cFc region of claim 5 .
7. The caninized antibody of claim 6 that further comprises a heavy chain complementary determining region 1 (VH CDR1) that comprises the amino acid sequence of SEQ ID NO: 29; a heavy chain complementary determining region 2 (VH CDR2) that comprises the amino acid sequence of SEQ ID NO: 33; a heavy chain complementary determining region 3 (VH CDR3) that comprises the amino acid sequence of SEQ ID NO: 38; a light chain complementary determining region 1 (VL CDR1) that comprises the amino acid sequence of SEQ ID NO: 15; a light chain complementary determining region 2 (VL CDR2) that comprises the amino acid sequence of SEQ ID NO: 18; and a light chain complementary determining region 3 (VL CDR3) that comprises the amino acid sequence of SEQ ID NO: 24.
8. The caninized antibody of claim 6 that further comprises a heavy chain complementary determining region 1 (VH CDR1) that comprises the amino acid sequence of SEQ ID NO: 27; a heavy chain complementary determining region 2 (VH CDR2) that comprises the amino acid sequence of SEQ ID NO: 31; a heavy chain complementary determining region 3 (VH CDR3) that comprises the amino acid sequence of SEQ ID NO: 36; a light chain complementary determining region 1 (VL CDR1) that comprises the amino acid sequence of SEQ ID NO: 13; a light chain complementary determining region 2 (VL CDR2) that comprises the amino acid sequence of SEQ ID NO: 19; and a light chain complementary determining region 3 (VL CDR3) that comprises the amino acid sequence of SEQ ID NO: 25.
9. The caninized antibody of claim 6 , wherein the cFc region further comprises a hinge region that comprises an amino acid sequence selected from the group consisting of SEQ ID NO: 109, SEQ ID NO: 110, SEQ ID NO: 111, and SEQ ID NO: 112.
10. The caninized antibody of claim 7 , wherein when bound to canine PD-1, said antibody binds to at least one amino acid residue within SEQ ID NO: 144; wherein the antibody binds canine PD-1 and blocks the binding of canine PD-1 to canine Programmed Death Ligand 1 (PD-L1).
11. The caninized antibody of claim 10 wherein said antibody binds to at least one amino acid residue within one or more amino acid sequences selected from the group consisting of SEQ ID NO: 138, SEQ ID NO: 139, SEQ ID NO: 140, SEQ ID NO: 141, of SEQ ID NO: 142, SEQ ID NO: 143, and SEQ ID NO: 145.
12. The caninized antibody of claim 11 , wherein said antibody binds to one or more amino acid residues selected from the group consisting of R 62 , R 69 , R 72 , R 75 , and R 90 of SEQ ID NO: 114.
13. A pharmaceutical composition comprising the antibody of claim 10 and a pharmaceutically acceptable carrier or diluent.
14. A caninized antibody comprising a heavy chain that comprises the amino acid sequence of SEQ ID NO: 44; wherein there is a proline residue at amino acid position 244, an alanine residue at amino acid position 271, an alanine residue at amino acid position 303, a glycine residue at amino acid position 304, a threonine residue at amino acid position 305, an alanine residue at amino acid position 333, and a proline residue at amino acid position 335; wherein said caninized antibody further comprises a canine light chain that comprises the amino acid sequence SEQ ID NO: 78.
