Abstract

There is disclosed compositions and methods relating to or derived from anti-ErbB3 antibodies. More specifically, there is disclosed fully human antibodies that bind ErbB3, ErbB3-binding fragments and derivatives of such antibodies, and ErbB3-binding polypeptides comprising such fragments. Further still, there is disclosed nucleic acids encoding such antibodies, antibody fragments and derivatives and polypeptides, cells comprising such polynucleotides, methods of making such antibodies, antibody fragments and derivatives and polypeptides, and methods of using such antibodies, antibody fragments and derivatives and polypeptides, including methods of treating or diagnosing subjects having ErbB3 related disorders or conditions, including various inflammatory disorders and various cancers.

Claims (12)

1. A recombinant fully human antibody that binds to an ErbB3 epitope, comprising a heavy chain variable domain comprising complementarity determining sequences (CDRs) as set forth in a heavy chain variable domain amino acid sequence selected from the group consisting of SEQ ID NO. 3, SEQ ID NO. 5, and SEQ ID NO. 7, and comprising a light chain variable domain comprising CDRs as set forth in a light chain variable domain amino acid sequence selected from the group consisting of SEQ ID NO. 4, SEQ ID NO. 6, and SEQ ID NO. 8, wherein the antibody is an IgG isotype.

2. The fully human antibody of claim 1 , wherein the antibody has a heavy chain/light chain variable domain combination selected from the group consisting of SEQ ID NO. 3/SEQ ID NO. 4, SEQ ID NO. 5/SEQ ID NO. 6, and SEQ ID NO. 7/SEQ ID NO. 8.

A fully human antibody Fab fragment comprising a heavy chain variable domain comprising CDRs as set forth in a heavy chain variable domain amino acid sequence selected from the group consisting of SEQ ID NO. 3, SEQ ID NO. 5, and SEQ ID NO. 7, and comprising a light chain variable domain comprising CDRs as set forth in a light chain variable domain amino acid sequence selected from the group consisting of SEQ ID NO. 4, SEQ ID NO. 6, and SEQ ID NO. 8.

4. The fully human antibody Fab fragment of claim 3 , wherein the antibody has a heavy chain/light chain variable domain combination selected from the group consisting of SEQ ID NO. 3/SEQ ID NO. 4, SEQ ID NO. 5/SEQ ID NO. 6, and SEQ ID NO. 7/SEQ ID NO. 8.

A single chain human antibody comprising a heavy chain variable domain, a light chain variable domain, and a peptide linker connecting the heavy chain variable domain and the light chain variable domain, wherein the heavy chain variable domain comprises CDRs as set forth in a heavy chain amino acid sequence selected from the group consisting of SEQ ID NO. 3, SEQ ID NO. 5, and SEQ ID NO. 7, and the light chain variable domain comprises CDRs as set forth in a light chain variable domain amino acid sequence selected from the group consisting of SEQ ID NO. 4, SEQ ID NO. 6, and SEQ ID NO. 8.

6. The fully human single chain antibody of claim 5 , wherein the single chain fully human antibody has a heavy chain/light chain variable domain combination selected from the group consisting of SEQ ID NO. 3/SEQ ID NO. 4, SEQ ID NO. 5/SEQ ID NO. 6, and SEQ ID NO. 7/SEQ ID NO. 8.

A method for treating a disease, comprising administering an effective amount of an anti-ErbB3 antigen binding protein selected from the group consisting of a recombinant fully human antibody of an IgG class that binds to an ErbB3 epitope with a binding affinity of at least 10 −6 M, a fully human antibody Fab fragment, and a single chain human antibody comprising a heavy chain variable domain, a light chain variable domain, and a peptide linker connecting the heavy chain variable domain and the light chain variable domain,

wherein the anti-ErbB3 antigen binding protein comprises a heavy chain variable domain comprising CDRs as set forth in a heavy chain variable domain amino acid sequence selected from the group consisting of SEQ ID NO. 3, SEQ ID NO. 5, and SEQ ID NO. 7, and comprises a light chain variable domain comprising CDRs as set forth in a light chain variable domain amino acid sequence selected from the group consisting of SEQ ID NO. 4, SEQ ID NO. 6, and SEQ ID NO. 8,

wherein the disease is cancer or an autoimmune disease.

8. The method of claim 7 , wherein the antigen binding protein comprises a heavy chain/light chain variable domain combination selected from the group consisting of SEQ ID NO. 3/SEQ ID NO. 4, SEQ ID NO. 5/SEQ ID NO. 6, and SEQ ID NO. 7/SEQ ID NO. 8.

9. The method of claim 7 , wherein the cancer is an ErbB3-positive cancer.

10. The method of claim 7 , wherein the broad the cancer is selected from the group consisting of carcinoma, prostate cancer, non-small cell lung cancer, breast cancer, endometrial cancer, ovarian cancer, gastric cancers, head and neck cancers, and colon cancer.
