Abstract

A novel combination comprising the VEGFR inhibitor 5-[[4-[(2,3-dimethyl-2H-indazol-6-yl)methylamino]-2-pyrimidinyl]amino]-2-methylbenzenesulfonamide, or a pharmaceutically acceptable salt thereof, with a B-Raf inhibitor, particularly N-{3-[5-(2-Amino-4-pyrimidinyl)-2-(1,1-dimethylethyl)-1,3-thiazol-4-yl]-2-fluorophenyl}-2,6-difluorobenzenesulfonamide or a pharmaceutically acceptable salt thereof, pharmaceutical compositions comprising the same and methods of using such combinations and compositions in the treatment of conditions in which the inhibition of VEGFR and/or B-Raf is beneficial.

Claims (18)

1 . A combination comprising:

(i) a compound of formula (I)

or a pharmaceutically acceptable salt thereof; and

(ii) a compound of formula (II)

or a pharmaceutically acceptable salt thereof.

2 . A combination according to claim 1 wherein compound (i) is in the form of the monohydrochloride salt and the compound (II) is in the form of the methanesulfonate salt.

3 . A combination kit comprising a combination according claims 1 to 2 together with a pharmaceutically acceptable carrier or carriers.

4 . Use of a combination according to any of claims 1 to 2 in the manufacture of a medicament for the treatment of cancer.

5 . A method of treating a susceptible cancer in a human in need thereof which comprises the administration of therapeutically effective amounts of

(i) a compound of formula (I)

or a pharmaceutically acceptable salt thereof; and

(ii) a compound of formula (II)

or a pharmaceutically acceptable salt thereof.

The method of claim 5 , wherein the susceptible cancer is selected from both primary and metastatic forms of head and neck, breast cancer, inflammatory breast cancer, lung cancer, non-small cell lung cancer (NSCLC), colon cancer, prostate cancers, primary CNS tumors such as gliomas, glioblastomas, astrocytomas and ependymomas, and secondary CNS tumors (i.e., metastases to the central nervous system of tumors originating outside of the central nervous system), Bannayan-Zonana syndrome, Cowden disease, Lhermitte-Duclos disease, Wilm's tumor, Ewing's sarcoma, Rhabdomyosarcoma, ependymoma, medulloblastoma, colorectal cancer, renal cancer, kidney cancer, liver, melanoma, ovarian cancer, pancreatic, sarcoma, osteosarcoma, giant cell tumor of bone, thyroid cancer (including papillary thyroid cancer), lymphoblastic T cell leukemia, Chronic myelogenous leukemia, Chronic lymphocytic leukemia, Hairy-cell leukemia, acute lymphoblastic leukemia, acute myelogenous leukemia (AML), Chronic neutrophilic leukemia, plasmacytoma, Immunoblastic large cell leukemia, Mantle cell leukemia, megakaryoblastic leukemia, multiple myeloma, acute megakaryocytic leukemia, promyelocytc leukemia, erythroleukemia, malignant lymphoma, hodgkins lymphoma, non-hodgkins lymphoma, lymphoblastic T cell lymphoma, Burkitt's lymphoma, follicular lymphoma, neuroblastoma, bladder cancer, urothelial cancer, vulval cancer, cervical cancer, endometrial cancer, mesothelioma, esophageal cancer, salivary gland cancer, hepatocellular cancer, gastric cancer, nasopharangeal cancer, buccal cancer, cancer of the mouth, GIST (gastrointestinal stromal tumor), Barret's adenocarcinoma; billiary tract carcinomas; cholangiocarcinoma; myelodysplastic syndromes, pituitary adenoma, and testicular cancer.

7 . The method of claim 5 , wherein the susceptible cancer is selected from melanoma, colorectal cancer, ovarian cancer, papillary thyroid cancer, and cholangiocarcinoma.

8 . A combination according to claims 1 to 2 for use in therapy.

9 . A combination according to claims 1 to 2 for use in treating cancer.

10 . A pharmaceutical composition comprising a combination according to claims 1 to 2 together with a pharmaceutically acceptable diluent or carrier.
