Bipyrazole derivatives as JAK inhibitors
View Patent ↗The present invention provides compounds of Formula I: or pharmaceutically acceptable salts thereof, as well as their compositions and methods of use, that inhibit the activity of Janus kinase (JAK) and are useful in the treatment of diseases related to the activity of JAK including, for example, inflammatory disorders, autoimmune disorders, cancer, and other diseases.
1. A compound which is 4-[3-(cyanomethyl)-3-(3′,5′-dimethyl-1H,1′H-4,4′-bipyrazol-1-yl)azetidin-1-yl]-2,5-difluoro-N-[(1S)-2,2,2-trifluoro-1-methylethyl]benzamide, or a pharmaceutically acceptable salt thereof.
2. A composition comprising a compound or salt according to claim 1 and a pharmaceutically acceptable carrier.
3. A method of inhibiting an activity of JAK1 comprising contacting JAK1 with a compound or salt according to claim 1 .
4. The method according to claim 3 , wherein said compound or salt is selective for JAK1 over JAK2.
5. A method of treating a solid tumor in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of a compound or salt according to claim 1 .
6. A method of treating prostate cancer in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of a compound or salt according to claim 1 .
7. A method of treating lymphoma in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of a compound or salt according to claim 1 .
8. A method of treating polycythemia vera (PV) in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of a compound or salt according to claim 1 .
9. A method of treating myelofibrosis in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of a compound or salt according to claim 1 .
10. A method of treating primary myelofibrosis (PMF) in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of a compound or salt according to claim 1 .
11. A method of treating post polycythemia vera myelofibrosis (Post-PV MF) in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of a compound or salt according to claim 1 .
12. A method of treating post-essential thrombocythemia myelofibrosis (Post-ET MF) in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of a compound or salt according to claim 1 .
13. A method of treating a myelodysplastic syndrome (MDS) in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of a compound or salt according to claim 1 .
14. The method according to claim 13 , wherein said myelodysplastic syndrome is refractory cytopenia with unilineage dysplasia (RCUD).
15. A method of treating renal cancer in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of a compound or salt according to claim 1 .
16. A method of treating hepatic cancer in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of a compound or salt according to claim 1 .
17. A method of treating breast cancer in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of a compound or salt according to claim 1 .
18. A method of treating lung cancer in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of a compound or salt according to claim 1 .
19. A method of treating thyroid cancer in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of a compound or salt according to claim 1 .
20. A method of treating Kaposi's sarcoma in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of a compound or salt according to claim 1 .
21. A method of treating Castleman's disease in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of a compound or salt according to claim 1 .
22. A method of treating pancreatic cancer in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of a compound or salt according to claim 1 .
23. A method of treating leukemia in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of a compound or salt according to claim 1 .
24. A method of treating multiple myeloma in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of a compound or salt according to claim 1 .
25. A method of treating essential thrombocythemia (ET) in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of a compound or salt according to claim 1 .
26. A method of treating chronic myelogenous leukemia (CML) in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of a compound or salt according to claim 1 .
27. A method of treating chronic myelomonocytic leukemia (CMML) in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of a compound or salt according to claim 1 .
28. A method of treating hypereosinophilic syndrome (HES) in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of a compound or salt according to claim 1 .
29. A method of treating idiopathic myelofibrosis (IMF) in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of a compound or salt according to claim 1 .
30. A method of treating systemic mast cell disease (SMCD) in a patient in need thereof, comprising administering to said patient a therapeutically effective amount of a compound or salt according to claim 1 .
31. The method according to claim 13 , wherein said myelodysplastic syndrome is refractory anemia with ring sideroblasts (RARS).
32. The method according to claim 13 , wherein said myelodysplastic syndrome is refractory cytopenia with multilineage dysplasia.
33. The method according to claim 13 , wherein said myelodysplastic syndrome is refractory anemia with excess blasts-1 (RAEB-1).
34. The method according to claim 13 , wherein said myelodysplastic syndrome is refractory anemia with excess blasts-2 (RAEB-2).
35. The method according to claim 13 , wherein said myelodysplastic syndrome is myelodysplastic syndrome.
36. The method according to claim 13 , wherein said myelodysplastic syndrome is unclassified (MDS-U).
37. The method according to claim 13 , wherein said myelodysplastic syndrome is myelodysplastic syndrome associated with isolated del(5q).
