Abstract

The present disclosure relates to certain polypeptides derived from prokaryotic DGT enzymes, and nucleic acids useful in encoding the same.

Claims (28)

1. An isolate nucleic acid molecule comprising:

a polynucleotide operably linked to a heterologous promoter, wherein the polynucleotide encodes a 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) linked to a chloroplast transit peptide (CTP), wherein the EPSPS is at least 90% identical to SEQ ID NO 1, and wherein the EPSPS comprises an alanine at the position corresponding to position 84 of SEQ ID NO 1.

2. The nucleic acid molecule of claim 1 , wherein the first methionine residue of the 5-enolpyruvylshikimate 3-phosphate synthase is replaced by the CTP.

3. A vector comprising the nucleic acid molecule of claim 1 .

4. The vector of claim 3 , comprising a further polynucleotide encoding a heterologous polypeptide.

5. The vector of claim 3 , wherein the vector is a plant transformation vector.

6. A host cell that contains the nucleic acid molecule of claim 1 , wherein the host cell is a bacterial cell or a plant cell.

7. The vector of claim 5 , wherein the promoter is an AtUbi10 promoter.

8. The host cell of claim 6 , wherein the host cell is a plant cell.

9. A transgenic plant material comprising the plant cell of claim 8 , wherein the plant material is a plant part, plant organ, or plant seed.

10. The transgenic plant material of claim 9 , wherein the plant material is tolerant to glyphosate, when compared to a wild-type plant of the same species that does not comprise the polynucleotide operably linked to the heterologous promoter.

11. The transgenic plant material of claim 9 , wherein the transgenic plant material is resistant to an herbicide selected from the group consisting of glufosinate; 2,4-dichlorophenoxyacetic acid (2,4-D); and an herbicide that inhibits acetolactate synthase (ALS).

12. A transgenic tissue culture of regenerable cells produced from the transgenic plant material of claim 9 .

13. A transgenic protoplast produced from the transgenic plant material of claim 9 .

14. The transgenic tissue culture of claim 12 , wherein the regenerable cells are produced from a tissue type selected from the group consisting of leaves, pollen, embryos, cotyledons, hypocotyls, meristematic cells, roots, root tips, anthers, flowers, stems and pods.

15. A plant regenerated from the tissue culture of claim 12 , wherein the plant is resistant to glyphosate.

16. A method of generating a plant, plant part, plant organ, plant seed, or plant cell resistant to glyphosate, the method comprising:

transforming the plant, plant part, plant organ, plant seed, or plant cell with the nucleic acid molecule of claim 1 ; and

expressing the polypeptide encoded by the polynucleotide, thereby generating the plant, plant part, plant organ, or plant cell resistant to glyphosate.

17. A method for controlling weeds in an area under cultivation containing herbicide-resistant plants, the method comprising:

planting a plant or a plant seed comprising the nucleic acid molecule of claim 1 in the area under cultivation; and

applying to the area under cultivation a sufficient amount of herbicide to give at least 85% control of a weed species in the area under cultivation.

18. The method according to claim 17 , wherein the herbicide is glyphosate.

19. The method according to claim 17 , wherein the nucleic acid molecule comprises a second polynucleotide encoding a heterologous polypeptide.

20. The method according to claim 19 , wherein the second polynucleotide is α-ketoglutarate-dependent dioxygenase 1 (aad-1) or α-ketoglutarate-dependent dioxygenase 12 (aad-12).

21. A plant having stably integrated into its genome the nucleic acid molecule of claim 1 .

The plant of claim 21 , wherein the plant is selected from the group consisting of wheat, corn, soybean, tobacco, brachiaria, rice, millet, barley, tomato, apple, pear, strawberry, orange, alfalfa, cotton, carrot, potato, sugar beets, yam, lettuce, spinach, petunia, rose, chrysanthemum, turf grass, pine, fir, spruce, heavy metal accumulating plants, sunflower, safflower, rapeseed, and Arabidopsis.

23. The plant of claim 21 , wherein the plant is a species selected from the group consisting of genera Asparagus, Avena, Brachiaria, Brassica, Citrus, Citrullus, Capsicum, Cucurbita, Daucus, Erigeron, Glycine, Gossypium, Hordeum, Helianthus, Lactuca, Lolium, Lycopersicon, Malts, Manihot, Nicotiana, Orychophragmus, Oryza, Persea, Phaseolus, Pisum, Pyrus, Prunus, Raphanus, Secale, Solanum, Sorghum, Triticum, Vitis, Vigna , and Zea.
