Assay for methoxetamine
View Patent ↗Components for enabling immunodetection of methoxetamine are described including immunogens, haptens, antibodies and kits.
1. A polyclonal antibody raised against an immunogen having the structure of:
wherein KLH is keyhole limpet hemocyanin,
wherein the polyclonal antibody binds to methoxetamine and N-desethylmethoxetamine, wherein the polyclonal antibody has a cross-reactivity of 24% for N-desmethylmethoxetamine, when compared to 100% cross-reactivity for methoxetamine and shows a cross-reactivity of less than 5.0% for each of ketamine, norketamine, phencyclidine, tramadol and tilidine, when compared to 100% cross-reactivity for methoxetamine; and
wherein said cross reactivity is measured by using N-(5-carboxypentyl)-N-desethylmethoxetamine-HRP as a detecting agent, wherein HRP is horseradish peroxidase.
2. The polyclonal antibody of claim 1 which has an IC 50 of <1.00 ng/ml for methoxetamine.
3. The polyclonal antibody of claim 1 , which has been purified.
4. A method of detecting or determining methoxetamine in a sample comprising:
i) contacting the sample with a detecting agent and the polyclonal antibody of claim 1 ; and
ii) detecting or determining the amount of the detecting agent bound to the antibody.
5. The method of claim 4 in which the detecting agent is N-(5-carboxypentyl)-N-desethylmethoxetamine-HRP.
6. The method of claim 5 , wherein the presence of the detecting agent is detected or determined by a colour change in response to reaction of HRP with a substrate.
7. The method of claim 6 , wherein the colour change is detected or determined by reading the absorbance at 450 nm.
8. A kit comprising the polyclonal antibody of claim 1 .
9. The kit of claim 8 , further comprising a detecting agent, wherein the detecting agent N-(5-carboxypentyl)-N-desethylmethoxetamine-HRP.
