Abstract

The invention is directed to the removal of serum gal-3 from circulation by plasmapheresis, comprising at least in part donor apheresis, using gal-3 binding agents in either a fixed bed, or in a form easily removed, such as by being complexed with magnetic particles. This method, on its own, brings a sharp reduction and relief from the inflammation and fibroses that can be induced by circulating gal-3. The process may be combined with the administration of gal-3 binding agents, such as modified citrus pectin, to further lower unbound gal-3 levels, to the point where gal-3 in the tissues may be addressed. This method may also be combined with removal of TNF receptors to provide an effective treatment for cancer.

Claims (13)

1. A method of treating a disease characterized by a tumor in a mammal, comprising:

Selectively removing an amount of galectin-3 (gal-3) from blood from said mammal and selectively removing an amount of soluble tumor necrosis factor (TNF) receptors from said blood from said mammal; and

Reintroducing said blood to said mammal, such that said mammal's blood exhibits a level of gal-3 and soluble TNF receptors following said treatment lower than said blood prior to said treatment.

The method of claim 1 , wherein said gal-3 and TNF receptors are removed by apheresis, wherein said blood is withdrawn from said mammal and directed through modules wherein said blood comes in contact with agents which bind gal-3, and wherein said blood is directed through modules wherein said blood comes in contact with agents that bind TNF receptors, and thereafter said blood is returned to said mammal.

3. The method of claim 1 , wherein said agents which bind gal-3 and TNF receptors are antibodies, antibody fragments and mixtures thereof which bind, respectively, gal-3 and soluble TNF receptors.

4. The method of claim 1 , wherein said soluble TNF receptors comprise R-1, R-2 or a combination of R-1 and R-2 TNF receptors.

5. The method of claim 1 , wherein at least ten percent (10%) of gal-3in the blood of said mammal is removed.

The method of claim 1 , further comprising administering to said mammal an agent selected from the group consisting of a pharmaceutical agent, a chemotherapeutic agent, or a combination thereof, wherein the efficacy of said agent is enhanced by the reduction in the reintroduced blood of said mammal of gal-3, soluble TNF receptors or both.

7. A composition of matter comprising blood withdrawn from a mammal, wherein said blood has been treated to selectively reduce the level of gal-3 and soluble TNF receptors below the level ordinarily present in said mammal in the absence of said treatment.

8. The composition of matter of claim 7 , wherein the concentration of gal-3 in said treated blood is at least 10% lower than gal-3 concentration in blood of said mammal prior to said blood having been treated.

The composition of matter of claim 7 , wherein said treated blood further comprises at least one agent added to said blood following its withdrawal from said mammal selected from the group consisting of a pharmaceutical agent, a chemotherapeutic agent, or a combination thereof, wherein the efficacy of said agent is enhanced by the reduction in the reintroduced blood of said mammal of the presence of gal-3, soluble TNF receptors or both.

10. The composition of matter of claim 9 , wherein said agent is a chemotherapeutic agent.

11. The composition of claim 7 , wherein said composition does not comprise whole cells.
