Abstract

Various fenofibrate dosage forms contain a plurality of beads or particles, where the beads or particles include a pharmaceutical composition comprising fenofibrate; from 0.3% to 10% by weight of the beads or particles of a surfactant; and from about 5% to about 15% by weight of the beads or particles of a water soluble or water dispersible cellulosic binder. The mass ratio of the drug to the binder in the dosage form is between about 3.5:1 and 4.5:1; and the dosage form produces a first Cmax in vivo that is between about 10% and about 50% higher than a comparative Cmax produced by a comparative dosage form. The comparative dosage form comprises the drug and the binder in a ratio of between about 5:1 and 15:1. The dosage form may contain a plurality of beads or particles, each of the beads or particles including a pharmaceutical composition containing from about 20% to about 55% by weight of fenofibrate; from 0.3% to 10% by weight of a surfactant; and from about 5% to about 15% by weight of a water soluble or water dispersible cellulosic binder.

Claims (55)

1. A dosage form comprising a plurality of beads or particles, each of said beads or particles comprising:

from 25% to 55% by weight of a drug selected from the group consisting of fenofibric acid, pharmaceutically acceptable salts thereof, pharmaceutically acceptable esters thereof, and prodrugs thereof, substantially all of said drug being incorporated in said plurality of beads or particles;

an effective amount of a surfactant; and

from about 5% to about 15% by weight of a water soluble or water dispersible cellulosic binder;

wherein the mass ratio of said drug to said binder in said dosage form is between about 3.5:1and 4.5:1.

2. The dosage form according to claim 1 , wherein each of said beads or particles comprises an inert core; and a pharmaceutical composition comprising said drug, said surfactant; and said cellulosic binder;

wherein said pharmaceutical composition is coated on said inert core.

3. The dosage form according to claim 1 , wherein:

each of said beads or particles comprises 45% to about 55% by weight of said drug.

4. The dosage form according to claim 1 , wherein:

each of said beads or particles comprises 25% to about 35% by weight of said drug.

5. The dosage form according to claim 3 , wherein:

said dosage form comprises 40 to 200 mg fenofibrate.

6. The dosage form according to claim 3 , wherein:

said dosage form and said comparative dosage form each comprise 40 to 200 mg fenofibrate.

7. A dosage form comprising a plurality of beads or particles, said beads or particles each comprising a pharmaceutical composition comprising:

from about 20% to about 55% by weight of said beads or particles of a micronized drug selected from the group consisting of fenofibric acid, pharmaceutically acceptable salts thereof, pharmaceutically acceptable esters thereof, and prodrugs thereof;

from 0.3% to 10% by weight of said beads or particles of a surfactant; and

from about 5% to about 15% by weight of said beads or particles of a water soluble or water dispersible cellulosic binder.

8. The dosage form according to claim 7 , wherein:

said pharmaceutical composition comprises 45% to about 55% by weight of said drug.

9. The dosage form according to claim 7 , wherein:

said pharmaceutical composition comprises 25% to about 35% by weight of said drug.

10. The dosage form according to claim 7 , wherein:

said dosage form comprises 40 to 200 mg fenofibrate.

11. A dosage form comprising a plurality of beads or particles, said beads or particles each comprising an inert core with a particle size of 20 to 50 mesh and a pharmaceutical composition coated on said inert core;

said pharmaceutical composition comprising:

from about 20% to about 55% by weight of a drug selected from the group consisting of fenofibric acid, pharmaceutically acceptable salts thereof, pharmaceutically acceptable esters thereof, and prodrugs thereof;

from 0.3% to 10% by weight of said beads or particles of a surfactant; and

from about 5% to about 15% by weight of a water soluble or water dispersible cellulosic binder;

wherein the mass ratio of said drug to said binder in said dosage form is between about 3.5:1and 4.5:1.

12. A method of reducing a cholesterol level and/or increasing high-density lipoprotein levels in a patient at risk of cardiovascular disease, comprising administering a dosage form to said patient;

said dosage form comprising a plurality of beads or particles, said beads or particles each comprising:

from 20% to 55% by weight of a drug selected from the group consisting of fenofibric acid, pharmaceutically acceptable salts thereof, pharmaceutically acceptable esters thereof, and prodrugs thereof;

from 0.3% to 10% by weight of a surfactant; and

from 5% to 15% by weight of a water soluble or water dispersible cellulosic binder;

wherein the mass ratio of said drug to said binder in said dosage form is between 3.5:1 and 4.5:1.

13. The method of claim 12 , wherein said dosage form is administered in conjunction with a statin.

14. The dosage form according to claim 11 , wherein said dosage form is further characterized by least one of the following conditions:

a) said drug is micronized; and

b) said surfactant is present in an amount of between about 0.5% by weight and about 5% by weight.

15. The dosage form according to claim 14 , wherein said dosage form is further characterized in that said inert core has a mesh size of 20 to 25 mesh; and

said surfactant is present in an amount of between about 0.5% by weight and about 3% by weight.

16. The dosage form according to claim 14 , wherein said dosage form is further characterized in that said inert core has a mesh size of 35 to 45 mesh; and

said surfactant is present in an amount of between about 0.5% by weight and about 3% by weight.

17. The dosage form according to claim 14 , wherein:

said micronized drug has a weight average particle size of between 1 and 15 microns, or

said micronized drug has a particle size range such that at least 99% of drug particles have a particle size of <50 microns; at least 90% of drug particles have a particle size of <15 microns; and no more than 10% of drug particles have a particle size of <1 micron.

18. The method according to claim 12 , wherein said dosage form is further characterized by least one of the following conditions:

a) said drug is micronized; and

b) said surfactant is present in an amount of between about 0.5% by weight and about 5% by weight.

19. The method according to claim 12 , wherein said dosage form is further characterized in that said beads or particles each comprise an inert core with a particle size of 20 to 50 mesh, said pharmaceutical composition being coated on said inert core.

20. The method according to claim 18 , wherein said an inert core has a particle size of 20 to 25 mesh.

21. The method according to claim 18 , wherein said an inert core has a particle size of 35 to 45 mesh.

22. The dosage form according to claim 11 , wherein the dosage form comprises the plurality of beads or particles, each bead or particle consisting of the inert core and the pharmaceutical composition coated on the inert core.
